摘要
目的:探讨中药复方养肝活血汤对大鼠酒精性肝病(Alcoholic Liver Disease,ALD,简称酒精肝)的作用及其机制。方法:构建大鼠酒精肝模型,并检测不同组别中(空白对照组,模型对照组,养肝活血汤组,硫普罗宁钠组)中血清生化指标谷草转氨酶(AST),谷丙转氨酶(ALT)和肝脏中B淋巴细胞瘤-2基因(Bcl-2)和Bax蛋白水平的变化。结果:本研究成功构建了大鼠酒精肝模型,并发现当用阳性药硫普罗宁钠和养肝活血汤对大鼠的酒精肝加以治疗时,AST,ALT均显著下降。说明养肝活血汤对酒精肝有明显的疗效。为了进一步研究养肝活血汤的作用,本研究检测了大鼠肝细胞中的Bcl-2和Bax,同样证明了养肝活血汤对大鼠肝细胞的凋亡有明显的抑制作用。结论:因此本研究对于酒精肝的治疗,特别是中药养肝活血汤对酒精肝的治疗奠定了基础,为进一步的临床研究提供了实验室数据。
Objective:To study the effect and mechanism of the Yanggan Huoxue decoction(养肝活血汤)on alcoholic liver in rats.Methods:The model rats with alcoholic liver were constructed,serum biochemical indexes,such as AST,ALT and Bcl-2 and Bax in the liver were detected in different groups(blank control group,model control group,the Yanggan Huoxue decoction group,Tiopronin sodium group).Results:The model rats with alcoholic liver were successfully constructed,and the AST and ALT were significantly decreased when the positive medicines,tiprop sodium and no the Yanggan Huoxue decoction,were used to treat the alcoholic liver in rats.It shows that the Yanggan Huoxue decoction had obvious curative effect on alcoholic liver.In order to further study the effect of the Yanggan Huoxue decoction,the Bcl-2 and BAX in the rat liver cells were detected.The Yanggan Huoxue decoction had obvious inhibitory effect on the apoptosis of liver cells was proved.Conclusion:Therefore,this study proved that the Yanggan Huoxue decoction was effective on alcoholic liver,and provides laboratory data for further clinical study.
