摘要
目的采用HPLC-FT-ICR MS技术研究栀子大黄汤在大鼠胆汁中的原形成分及其代谢产物。方法色谱分离采用Shim-pack XR-ODS II色谱柱(75 mm×3.0 mm,2.2μm),流动相:体积分数为0.1%的甲酸水溶液-乙腈,梯度洗脱;质谱采用电喷雾电离源,在正、负离子模式下检测;对大鼠空白胆汁样品和给予栀子大黄汤后的胆汁样品进行分析,获取各成分的保留时间、精确质量数、二级碎片等信息,对栀子大黄汤在大鼠胆汁中的原形成分及其代谢产物进行鉴定。结果在大鼠胆汁中共检测到44个与栀子大黄汤相关的化学成分,包括30个原形成分和14个代谢产物。结论为阐明栀子大黄汤化学成分在大鼠体内的代谢和排泄过程提供了实验依据。
Objective To investigate the prototype components and metabolites of Zhi-Zi-Da-Huang decoction in rat bile after oral administration by HPLC-FT-ICR MS.Methods The chromatographic separation was performed using a Shim-pack XR-ODS II column(75 mm×3.0 mm,2.2μm).The elution gradient of mobile phase consisting of 0.1%formic acid-water and acetonitrile was used.The electrospray ionization(ESI)source was used and the data were acquired in both positive and negative ion modes for MS detection.By analysis of the blank bile and drug-dosed bile samples in rats,the information such as retention time,accurate mass and ion fragmentation was obtained,and the prototype components and metabolites of Zhi-Zi-Da-Huang decoction in rat bile were identified.Results A total of 44 constituents related to Zhi-Zi-Da-Huang decoction,including 30 prototype components and 14 metabolites were tentatively identified in rat bile.Conclusion This study provides the experimental basis for an insight into the metabolism and excretion of chemical components of Zhi-Zi-Da-Huang decoction in rats.
