摘要
目的:探讨复方片仔癀肝宝(GB)对化学性肝损伤的保护作用。方法:将60只SD大鼠按照随机数字表法分为正常对照组、模型组、GB(高、中、低)剂量组、西利宾胺组各10只。正常对照组腹腔注射橄榄油溶液1mL/kg,其余各组腹腔注射50%CCl4橄榄油溶液1 mL/kg,连续8周,2次/周。于造模第4周开始给药,模型组和正常对照组给予生理盐水;西利宾胺组给予西利宾胺3.3 mg/kg;GB低、中、高剂量组分别按GB 150、300、600 mg/kg给药,1次/d,连续4周。末次给药后禁食24 h,检测MDA、SOD、GSH和TBARs,HE染色观察肝组织病理学的影响,Real-time PCR和Western Blot检测肝组织CYP2E1的表达。结果:与模型组比较,各用药组中MDA、TBARs的活性均降低,SOD、GSH的活性升高,差异均有统计学意义(P<0.05);肝脏病理切片中,各用药组与模型组比较,肝损伤均有减轻的趋势,均下调CYP2E1的表达,差异具有统计学意义(P<0.05)。结论:GB能显著减轻CCl4诱导的化学性肝损伤,可能与GB的抗氧化应激有关。
Objective:To explore the hepatic protection of Pien Tze Huang Gan Bao (GB) on chemical liver injury. Methods: Sixty SD rats were randomly divided into control group, model group, silymarin group, and GB-low dose group, GB-middle dose group, GB-high dose group with ten rats per group. The control group received olive oil ( 1 mL/kg) intraperitoneally, rest of the groups received CC14 ( 1 mL/kg) intraperitoneally, twice per week for 8 weeks. From the fourth to the eighth week, the model group and the control group of rats were orally administrated saline, while the silymarin group of rats were administrated silymarin (3.3 mg/kg), and GB-Low group, GB-middle group, and GB-high group of rats were administrated 150 mg/kg, 300 mg/kg, and 600 mg/kg of GB, re- spectively. Twenty-four hours after the last dose, MDA, SOD, GSH and TBARs were detected; the histopathological changes in liver were observed by HE staining, and CYP2E1 gene transcription was determined by Real-time PCR. Results:The contents of MDA and TBARs were decreased in both silymarin and GB treated group of rats and with increasing the activity of SOD and GSH (P〈0.05 ); Compared with model, silymain and GB ameliorated the liver injuried. Furthermore, silymarin and GB down-regulated the expression of CYP2E1 (P〈0.05). Conclusion :GB could ameliorate the chemical liver injury, and it maybe correlate with the anti-oxidant activity of GB.
