摘要
目的:探讨人参皂苷Rg1对人脐静脉内皮细胞(HUVECs)基质金属蛋白酶-9(MMP-9)及血管内皮生成因子(VEGF)表达的影响,为临床治疗自身免疫性疾病提供理论及实验依据。方法:以TNF-α10 ng/ml刺激HUVECs,在上述条件中加入不同浓度Rg1(0,0.4,4及40 mmol/L),培养24 h后,分别用实时荧光定量PCR(real-time PCR)及免疫印迹实验(western Blot)检测MMP-9及VEGF的表达。结果:在基因水平,Rg1能抑制由TNF-α诱导HUVECs的MMP-9和VEGF的表达;在蛋白水平,Rg1能显著下调TNF-α诱导HUVECs的MMP-9和VEGF的表达。结论:Rg1通过抑制由TNF-α诱导HUVECs的MMP-9和VEGF的表达,从而在RA疾病治疗过程中发挥作用。
Objective: To investigate the regulatory effect of Rg1 on expressions of MMP- 9 and VEGF in human umbilical vein endothelial cells( HUVECs). Methods: Real- time fluorescent quantitative PCR and Western Blotting methods were used to detect the expressions of MMP- 9 and VEGF in presence or absence of Rg1 in human umbilical vein endothelial cells with the stimulation of TNF- α( 10 ng / m L). Results: The mRNA and protein expressions of MMP- 9 and VEGF were decreased in presence of Rg1( P 〈0. 05). Conclusion: Rg1 may inhibit the expressions of MMP- 9 and VEGF in human umbilical vein endothelial cells.
