摘要
目的:探讨参麦总皂苷抗血管生成作用,为临床辅助抗肿瘤分子机理提供理论依据。方法:提取参麦总皂苷,采用MTT法及刮痕法分析总皂苷对人脐静脉内皮细胞HUVECs的增殖和迁移抑制作用;通过荧光定量PCR方法考察总皂苷对人肝癌HepG2细胞株VEGF表达影响;最后,以鸡胚绒毛尿囊膜(CAM)在体模型对总皂苷抗血管生成作用进行评价。结果:MTT及刮痕实验结果表明,参麦总皂苷可剂量依赖性抑制HUVECs细胞增殖和迁移,其中600μg/mL浓度增殖抑制率达63.9%,150μg/mL处理48h,其迁移抑制率可达73.7%;荧光定量PCR结果表明,总皂苷可以抑制HepG2细胞分泌VEGF;此外,鸡胚绒毛尿囊膜模型证实,参麦总皂苷具有抗血管生成作用。结论:参麦总皂苷可以通过抑制内皮细胞增殖、迁移及抑制癌细胞分泌VEGF表达起到抗肿瘤血管生成作用。
Objective : To study the effect of total Saponins from Shenmai on anti - angiogenic, and provid a theory ba- sis for melocular mechanism of anti - tumor on clinical assistant. Methods : The total saponins was extracted from Shen- mai, MTT method and scratch test were employed to analysis the inhibitory effects of it on proliferation and migration of HUEVCs ; Effect of total Saponins on the expression of VEGF in HepG2 was examined with the fluorscence quantitative PCR. And in addition, CAM model was used to give an evaluation on the anti - angiogenesis effect of total Saponins. Re- sults:The results of MTT and scratch test showed that the total Saponins from Shenmai can inhibite the proliferation and migration of HUEVCs and the inhibition was a does - dependent. Among them, the inhibition ratio was 63.9% when the concentration at 600μg/ml, and at the concentration of 150μg/ml, the migration inhibitory rate was 73.7% at 48h post - treatment. The results of fluorscence quantitative PCR showed that the total Saponins can inhibit the secrete of VEDF in HepG2. Besides, the CAM model confirmed that total Saponins from Shenmai have the function to anti - angiogenesis. Conlution:The total Saponins from Shenmai produced the anti - angiogenesis effect by inhibit the proliferation and migra- tion of endothelial cell, and down - regulation the expression of VEGF in tumor cells.
