摘要
目的探讨丹红注射液对鸡绒毛尿囊膜(CAM)血管生成的影响。方法种蛋在(37.5±0.5)℃条件下孵育,第7天开窗暴露CAM建立模型。将60枚存活鸡胚随机分为生理盐水组(N组)、正常血清组(C组)、丹红注射液治疗组(T组)、血管内膜损伤组(I组)、血管内膜损伤后丹红注射液治疗组(IT组)以及血管内皮生成因子(VEGF,20μg/mL)组(V组),每组10枚鸡胚。新西兰大白兔腹主动脉用球囊导管损伤建立血管内膜损伤模型,丹红注射液治疗组及血管内膜损伤后丹红注射液治疗组于大白兔耳缘静脉注射丹红注射液2mL/kg,各组连续7d后取血清。第8天,在鸡胚CAM上放一直径为5mm的滤膜作为载体,分别加样5μL,每日1次,连续3d,第11天取CAM,数码相机拍照后平铺于载玻片上,计数载体周围血管数目及滤膜周围0.5cm范围内的血管分支点数并进行比较。结果比较CAM周围血管数目,T组(31.80±3.08)条与C组(28.40±3.75)条比较,血管以载体为中心呈辐辏状生长,血管总数明显增多,具有统计学意义;I组(47.30±3.27)条与C组相比较,载体周围血管数量明显增多,具有统计学意义;与IT组(47.60±3.89)条相比较,血管总数无统计学意义。结论含丹红注射液的兔血清能够明显促进CAM上血管生成,血管内膜损伤后的血清和内膜损伤丹红治疗组也分别强于血管内膜未损伤时。
Objective To investigate the angiogenesis effect of Danhong injection on the chick embryo chorioallantoic membrane(CAM) model.Methods Fertilized chick eggs were incubated and made a window in every egg on 7th day.Sixty survivors were randomly divided into 6 groups:Normal saline group(group N),normal rabbits serume group(group C),rabbits serum-containing Danhong injection(group T),serum of rabbit suffered with vascular intima injury(group I),serum-containing Danhong injection group suffered with vascular intima injury(group IT) and vascular endothelial growth factor group(VEGF,20μg/mL,group V).In group T and group IT Danhong injection 2 mL/kg was administered for seven days.On 8th day,one disc was placed on every CAM and 5 μL sample of each group was subtracted onto discs once a day for three days.The CAMs were harvested at 11th day and the number of blood vessel around the discs was counted.Results The proliferation of vessels in group T(31.80±3.08) and group I(47.30±3.27) was obviously higher than that in group C(28.40±3.75)respectively.Compared with group I,that in total number of vessels in the group IT(47.60±3.89) had no obviously difference.Conclusion Serum-containing Danhong injection could stimulate angiogenesis of CAM.The blood serum and serum-containing Danhong injection from vascular intima had more effective angiogenesis as comparison with normal serum.
