摘要 目的:建立黄芩的超高效液相色谱-电喷雾-飞行时间质谱(UPLC-ESI-TOF/MS)分析方法,研究黄芩的化学成分。方法:分别采集UPLC-TOF/MS全扫描质量色谱图和源内裂解的UPLC-TOF/MS色谱图,通过分子离子精确质量和同位素拟合度的分析确定分子式,在数据库中检索匹配的化合物,进而解析源内裂解的质谱图,从而对各峰进行鉴定或结构表征。结果:共鉴定或表征了黄芩药材中31个成分,均为黄酮类化合物。结论:该方法快速、灵敏、准确,可用于黄芩化学成分的分析,也为其它中药的成分分析提供了思路。 Objective : A method using ultra performance liquid chromatography - electrospray ionization - time of flight mass spectrometry (UPLC -ESI -TOF/MS) was developed to analyze the constituents in Scutellariae Radix. Methods:The full- scan UPLC -TOF/MS chromatogram and the chromatogram produced using in - source collision -induced dissociation were acquired, respectively. Through calculating accuracy mass and i- sotopic fit value of the molecular ion cluster, molecular formula was confirmed, and further the matched com- pound was researched in the established database composed of the known constituents in Scutellariae Radix. Finally, the peaks were identified or characterized structurally by elucidating the mass spectrum produced u- sing in - source collision - induced dissociation. Results : As a result, 31 compounds in Scutellariae Radix were detected, all of which were assigned to the flavonoids. Conclusion:It is concluded that this method is rapid, sensitive and accuracy, and can be used for analysis of the constituents in Scutellariae Radix. This study also provides a method to the analysis of other traditional Chinese medicines.
