摘要
目的:基于UHPLC-QTOF-MS法辨识藏药玛诺系汤三味颗粒中的药效物质基础。方法:玛诺系汤三味颗粒的甲醇:水(80∶20)提取液,用ZORBAX RRHD Eclipse Plus C_(18)色谱柱(3.0 mm×100 mm);流动相0.1%甲酸水-甲醇,梯度洗脱;体积流量0.4 mL/min;进样量2μL;柱温40℃;质谱采用双喷雾(ESI)离子源,在正、负离子模式下采集数据,通过保留时间、精确分子离子峰和二级质谱裂解碎片,按分子量、同位素分布和保留时间辨识化合物。结果:共鉴定128个化合物,能够准确识别的成分包括木香烃内酯、木质素、宽筋藤苷B、鞣花酸等40多种。结论:该方法高效、准确、稳定、重复性好,可以快速辨识藏药复方制剂的药效成分,为藏药复方质控指标确定提供参考。
Objective:Based on UHPLC-QTOF-MS method to identify the pharmacodynamic material basis in Tibetan medicine Manuoxitang Sanwei Granules.Method:Methanol:water(80×20)extract of Manuoxitang Sanwei Granules,using ZORBAX RRHD Eclipse Plus C_(18)column(3.0 mm×100 mm);mobile phase 0.1%formic acid water-methanol,gradient elution;volume The flow rate is 0.4 mL/min;the injection volume is 2μL;the column temperature is 40℃;the mass spectrometry adopts the double spray(ESI)ion source,and the data is collected in the positive and negative ion modes,and the retention time,accurate molecular ion peaks and secondary mass spectrometry fragmentation Fragments,identify compounds by molecular weight,isotopic distribution and retention time.Results:A total of 128 compounds were identified,and more than 40 kinds of components that could be accurately identified included woody hydrocarbon lactones,lignin,quinoside B,and ellagic acid.Conclusion:The method is efficient,accurate,stable,and has good repeatability.It can quickly identify the medicinal components of Tibetan medicine compound preparations,and provide a reference for the determination of Tibetan medicine compound quality control indicators.
