基于UPLC-Q-Orbitrap HRMS技术的白毛银露梅化学成分研究 被引量:17

摘要 目的:应用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)技术鉴定白毛银露梅中的主要化学成分。方法:采用AccucoreaQ RP18(2.1 mm×150 mm, 2.6μm)色谱柱,以甲醇-0.1%甲酸水溶液为流动相进行梯度洗脱,流速0.3 mL/min,柱温25℃。质谱条件采用高效电喷雾离子源(HESI),在负离子模式下,一级全扫描/数据依赖型二级质谱扫描模式检测,分析鉴定白毛银露梅中的主要化学成分。结果:结合高分辨质谱提供的一级准分子离子峰和二级碎片信息,参照相关文献,以及对照品比对,共鉴定出36个化学成分,包括19个黄酮类成分、9个有机酸类成分、6个儿茶素类成分和2个香豆素类成分,均为首次报道,其中5个成分经对照品进行了验证。结论:该方法可快速分析鉴定白毛银露梅中的化学成分,为合理开发其药用及食用价值提供科学依据。 Objective: To identify the main chemical constituents of Potentilla glabra var.mandshurica based on ultra-performance liquid chromatography-quadrupole-orbitrap high resolution mass spectrometry(UPLC-Q-Orbitrap HRMS).Methods: The chromatographic separation was performed on an AccucoreaQ RP18(2.1 mm×150 mm, 2.6 μm)column, the mobile phase was methanol-0.1% formic acid solution with gradient elution.The flow rate was 0.3 mL/min, column temperature was 25 ℃.Under the condition of high efficiency electrospray ion source(HESI)and the negative ion mode, the primary full scan/data dependent secondary mass spectrometry scanning mode was used to detect and analyze the main chemical constituents in Potentilla glabra var.mandshurica.Results: Combined with the primary excimer ion peak and secondary fragment information provided by high resolution mass spectrometry, thirty-six components, including 19 flavonoids, 9 organic acids, 6 catechins and 2 coumarins, were identified by referring to relevant literature and comparing with reference materials, both were first reported, and five of them were confirmed by reference standards.Conclusion: The established method can analyze the components in Potentilla glabra var.mandshurica rapidly, which provides evidence for the exploration of medicinal and edible value of this plant reasonably.
