摘要
目的:应用高效液相色谱串联四级杆飞行时间质谱(HPLC-Q-TOF-MS/MS)技术对蒙药安神补心六味丸的化学成分进行分析鉴定和分类归属。方法:采用Diamonsil^(ⓒ)C_(18)(2)(250 mm×4.6 mm,5μm)色谱柱,以甲醇-0.05%甲酸水为流动相,梯度洗脱,流速0.5 mL/min,柱温40℃;采用电喷雾离子源,在正、负离子模式下进行数据采集,根据化合物的精确质荷比、二级质谱裂解碎片,结合对照品、文献数据的比对进行碎片裂解规律分析鉴定。结果:共鉴定出119个化合物,包括36个有机酸类、22个黄酮及其苷类、8个木脂素类、3个倍半萜内酯类、6个萜类、3个香豆素类、17个酚类、14个氨基酸类、4个醛类和6个其他类。结论:首次对蒙药安神补心六味丸的化学成分进行快速辨识,为其药效物质基础研究以及质量控制奠定了基础。
Objective:To identify and classify the chemical constituents of Mongolian medicine Anshen buxin liuwei pills by high performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF-MS/MS)technology.Methods:The analysis was performed on Diamonsil^(ⓒ)C_(18)(2)(250 mm×4.6 mm,5μm)column,the mobile phases was methanol and 0.05%formic acid solution with gradient elution,flow rate was 0.5 mL/min,column temperature was 40℃.Electrospray ion source was used to collect data under positive and negative ion mode.Fragments were analyzed and identified according to the precise mass charge ratio of the compounds,secondary mass spectrometry,and comparison of reference materials and literature data.Results:A total of 119 compounds were identified,including 36 organic acids,22 flavonoids and their glycosides,8 lignans,3 sesquiterpene lactones,6 terpenes,3 coumarins,17 phenols,14 amino acids,4 aldehydes and and 6 others.Conclusion:The chemical components of Mongolian medicine Anshen buxin liuwei pills are rapidly identified for the first time,which lays a foundation for the research of pharmacodynamic material basis and quality control.
