摘要
目的制备慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠模型,研究黄芩苷对其防治作用及可能机制。方法成年雄性Wistar大鼠25只,分为空白组(F组),阳性对照组(C组),大、中、小剂量黄芩苷防治组(T1组、T2组、T3组),采用反复熏香烟加气管内两次灌注脂多糖的方法制备COPD大鼠模型,并于制备模型过程中进行干预。干预结束后处死大鼠,观察各组大鼠肺组织HE染色病理改变;测定平均肺泡数(MAN);肺组织干湿重比(RDW)及肿瘤坏死因子α(TNF—α)免疫组织化学阳性表达的灰度值。结果造模成功。各组大鼠RDW:F组15.36±0.43;C组10.86±0.51;T1组13.86±0.44;T2组13.16±0.42;T3组12.24±0.36。各组大鼠肺组织MAN:F组99.00±5.92;C组40.00±5.93;T1组65.00±5.15;T2组62.00±6.32;T3组58.00±4.47。各组大鼠肺组织TNF-α免疫组织化学阳性表达灰度值:F组137.11±3.26;C组58.75±1.40;T1组113.92±4.62;T2组84.83±5.32;T3组65.43±5.85。结论黄芩苷干预COPD模型大鼠,可以改善大鼠肺部支气管炎、肺气肿的病理变化;提高其MAN、RDW降低TNF-α在肺组织的表达,显示对大鼠COPD有一定的防治作用。黄芩苷可能在防治COPD中有一定的前景。
Objective To study prevention and treatment effects of baicalin in model rats lung with chronic obstructive pulmonary disease(COPD) by COPD model rats. Methods According to the principle of random, 25 Wistar rats (male) were divided into five groups, free group (group F), model contrast group (group C),maximum dose baicalin,middle dose and minimum dose treatment groups (group T1 ,group T2, group T3 ). Model rats were established by intratracheal instillation of lipopolysaccharide twice and exposed to cigarette smoke. In the process of model establishment,baicalin was given for treatment. After treatment, the rats were killed to study the pathological change of lung tissue with HE staining in each group, and measure mean alveolar number (MAN), the ratio of dried lung weight to fresh lung weight (RDW), immunohistochemical stains were performed to detect the expression of TNF-α. Results The COPD rats model was successfully made. The RDW of group F was 15.36±0.43,group C 10.86±0. 51,group T1 13. 86± 0.44, group T213.16 ± 0.42, group T312.24 + 0.36. The MAN of group F was 99.00 ±5.92, group C 40.00 ± 5.93,group T1 65.00 ±5.15, group T2 62.00 ± 6.32, group T3 58. 00 ± 4.47, TNF-α immunohistochemical stains were performed,group F was 137.11±3.26,group C 58.75±1.40,group T1 113.92.00±4.62,group T2 84.83 ± 5.32,group T365.43 ±5.85. Conclusions Baicalin has some effects on COPD model rats. There is some outlook on baicalin on prevention and treatment of COPD.
