摘要
目的:采用烟熏联合气管内脂多糖(lipopolysaccharide,LPS)滴入方法建立慢性阻塞性肺疾病(chronic obstructive pulmo-nary disease,COPD)大鼠模型,探讨淫羊藿苷对COPD模型大鼠超氧化物歧化酶(superoxide dismutase,SOD)活力的影响。方法:将50只雄性SD大鼠随机分为5组,健康对照组(A组),不予处理;COPD模型组(B组)、淫羊藿苷组(C组)、泼尼松组(D组)、淫羊藿苷加泼尼松组(E组),建立大鼠COPD模型并分别给予蒸馏水(10 mL/kg)、淫羊藿苷(100 mg/kg)、泼尼松(5 mg/kg)、淫羊藿苷(50 mg/kg)加泼尼松(2.5 mg/kg)灌胃治疗。在光镜下观察大鼠肺组织病理改变,测定外周血和肺组织匀浆中SOD活力。结果:光镜下A组肺泡结构完整,肺泡壁无炎性细胞浸润;B组肺泡壁及肺间隔炎性细胞浸润,肺间隔断裂、融合,肺实质破坏,形成广泛性肺气肿;C组肺泡壁及肺间隔内炎性细胞浸润明显,肺泡腔扩大,部分出现融合,但肺实质结构破坏程度轻;D组肺组织病理改变近似A组;E组改变介于C组与D组之间,肺泡腔扩大,肺间隔断裂、融合不明显,肺组织炎性浸润明显轻于C组。与A组比较,B组肺组织匀浆及外周血SOD活力显著下降(P<0.01);C、D、E组肺组织匀浆SOD活力下降(P<0.01),外周血中SOD活力呈下降趋势,但差异无统计学意义(P>0.05)。与B组比较,C、E组肺组织匀浆及外周血SOD活力下降较轻(P<0.05);C、E组SOD活力下降趋势相似,差异无统计学意义(P>0.05)。结论:淫羊藿苷能减轻肺组织局部及外周血中的SOD活力的下降,减轻氧化应激对肺组织的损伤。
Objective:To observe the effect of icariin on superoxide dismutase(SOD) activity in a rat model of chronic obstruc- tive pulmonary disease(COPD) induced by cigarette smoking and lipopolysaccharide dripped to trachea. Methods:A total of 50 male adult Sprague-Dawley rats were randomly divided into five groups: control group (Group A), model group (Group B), icariin group (Group C), prednisone group (Group D), icariin and prednisone group(Group E). Rats in group A were bred normally. The rest of rats were placed into sealed boxes full of smoke to build rat model of COPD. Before smoking, rats were treated with distilled water at a dose of 10 mL/kg in Group B, icariin at a dose of 100 mg/kg in Group C, prednisone at a dose of 5 mg/kg in Group D, icariin at a dose of 50 mg/kg and prednisone at a dose of 2.5 mg/kg in Group E. The activity of SOD was detected by method of xanthine oxidase. Results: The activity of SOD decreased in serum and lung homogenate of Group B, compared with Group A (P〈0.01). Pretreatment with icariin or icariin and prednisone could alleviate the sharp decline in SOD activity, compared with Group B(P〈0.05). There were no significant differences between Group C and Group E(P〉0.05). Conclusions: Icariin can inhibit the decline in SOD activity in the lung tissues and peripheral blood, and thus alleviate oxidative stress damage in the lung tissues.
