摘要
目的探讨外源性肺表面活性物质(PS)对慢性阻塞性肺疾病(COPD)大鼠模型的预防保护作用。方法采用香烟熏吸加气道内注入脂多糖法建立大鼠COPD模型,并应用PS进行干预,观察其肺功能、动脉血气、肺组织病理学改变及PS相关蛋白A(SP-A)的表达情况。结果本文建立被动吸烟致肺损伤大鼠模型的肺组织病理学改变基本符合人类COPD组织病理学改变特点,应用PS干预后,肺组织的损伤减轻,肺功能改善,SP-A的表达增多。结论应用外源性PS进行干预可改善COPD大鼠肺功能,明显抑制气道炎症,减轻肺组织损伤,对COPD的发生起到预防保护作用。
[Objective] To investigate the protective and therapeutic effect of pulmonary surfactant in the pathogenesis of chronic obstructive pulmonary disease (COPD) in rats. [Methods] COPD animal model was established by smoke inhalations and intracheal instillations of lipopolysaccharide in Wistar rats. Lung function test, blood gas analysis, light microscopy and electromicroscopy observations were performed. The expression of surfactant protein A (SP-A) were observed by immunohistochemistry. [Results] Smoking for 6 weeks and instillations of lipopolysaccharide twice resulted in chronic bronchitis and emphysema. Administration of exogenous pulmonary surfactant for 4 times resulted in statistically significant inhibition of pulmonary injury. [Conclusion] Pulmonary surfactant may have a protective and therapeutic effect on COPD model of rats by alleviating airway inflammatory reaction, decreasing the destruction of alveolar and improving the lung function.
