摘要
以二至四年生西洋参根际土壤为试材,采用化学测定技术、高通量测序技术,研究了不同种植年限对西洋参根际土壤的理化性质及根际微生物群落多样性的影响,以期为西洋参微生物的促生研究及连作障碍的防治提供参考依据。结果表明:有机质、全磷、速效钾、全钾含量呈逐年减少的趋势,其中全磷、速效钾含量显著低于前一年。QX4根际土壤细菌、真菌群落的ACE指数、Chao1指数均显著高于其他2个年份,说明QX4物种丰富度最高。根际土壤细菌共注释到30个门、198个目、600个属,门水平上主要以变形菌门Proteobacteria、酸杆菌门Acidobacteriota为优势菌群,相对丰度共占58.49%~68.72%。根际土壤真菌共注释到14个门、91个目、343个属,门水平上优势菌门有子囊菌门Ascomycota、被孢霉门Mortierellomycota,相对丰度共占76.07%~80.30%;属水平上被孢霉属Mortierella在3个年份中为共有的优势菌群。关联分析发现,速效磷、全钾含量与鞘氨醇单胞菌属丰度呈显著正相关。
Taking the rhizosphere soil of Panax quinquefolius at 2 to 4 years old as the test material,the effects of different planting years on the physical and chemical properties of the rhizosphere soil and the diversity of rhizosphere microbial communities were studied by using chemical determination technology and high-throughput sequencing technology,in order to provide reference for the research on the growth promotion of microorganisms and the prevention and control of continuous cropping obstacles for P.quinquefolius.The results showed that the contents of organic matter,total phosphorus,available potassium and total potassium decreased year by year,and the contents of total phosphorus and available potassium were significantly lower than that of the previous year.ACE index and Chao1 index of rhizosphere soil bacterial and fungal communities in QX4 were significantly higher than those in the other two years,indicated that QX4 had the highest species richness.The bacteria communities in rhizosphere soil were identified as 30 phyla,198 orders and 600 genera.At the phyla level,the dominant bacteria were Proteobacteria and Acidobacteriota,with a relative abundance ranging from 58.49%to 68.72%.The fungal communities in rhizosphere soil were identified as 14 phyla,91 orders,and 343 genera.The dominant phyla were Ascomycota and Mortierellomycota,with a relative abundance ranging from 76.07%to 80.30%.At the generic level,Mortierella was the dominant bacterial group in all three years.There was a significant positive correlation between the content of available phosphorus and total potassium and relative abundance of Sphingomonas.
