摘要
目的运用网络药理学研究脾胃培源方君药药对白术-黄芪治疗胃癌的作用机制。方法使用TCMSP数据库检索白术、黄芪的有效活性成分与作用靶点,通过Genecards、Drugbank、PharmGKB、OMIM、TTD数据库检索胃癌疾病靶点,将预测的靶点与疾病靶点取交集获得交集基因,通过Cytoscape3.7.2搭建"药物-化合物-靶点"网络。利用String将交集基因导入并分析,搭建蛋白互作网络。基于Metascape进行GO生物过程与KEGG通路富集分析,依据结果绘制相应结果图,运用Cytoscape 3.7.2构建"靶点-通路"网络,研究其作用机制。结果"药物-化合物-靶点"网络包含20个化合物、168个靶点,关键化合物为槲皮素、山柰酚、芒柄花素等;蛋白互作网络关键靶点主要为AKT1、IL-6、TP53、VEGFA、TNF、JUN等。GO生物过程分析筛选到前20个生物过程,包括对有毒物质、无机物及脂多糖的反应、对损伤的反应、活性氧代谢、凋亡信号通路等;KEGG通路富集分析筛选得到12条与胃癌相关的通路,有癌症通路、IL-17、PI3K-Akt信号通路等。结论白术-黄芪药对可能是通过介导癌症通路、IL-17、PI3K-Akt等信号通路,肿瘤细胞对有毒物、无机物、脂多糖的反应,以及调控肿瘤细胞的损伤、氧代谢、增殖、凋亡等生物过程进而起到治疗胃癌的作用。
Objective Based on network pharmacology to study the mechanism of action of Spleen and Stomach Peiyuan Prescription on the treatment of gastric cancer by Atractylodes-Astragalus.Methods Using TCMSP database to search for the effective active ingredients and targets of Atractylodes and Astragalus,and search for gastric cancer disease targets through Genecards,Drugbank,PharmGKB,OMIM,TTD databases,and then take the intersection of the targets predicted by the active ingredients and disease targets obtain the intersection genes,and use Cytoscape 3.7.2 to construct a“drug-compound-target”network.Import the intersection genes into String and draw the PPI network.Using Metascape to perform GO biological process and KEGG pathway enrichment analysis,drawing GO bar graphs and KEGG bubble graphs based on the results,Using Cytoscape 3.7.2 to construct a“target-pathway”network,and study its mechanism of action.Results The“drug-compound-target”network contains 20 active compounds and 168 targets.The key compounds are quercetin,kaempferol,formononetin,etc;the key targets of the PPI network are mainly AKT1,IL-6,TP53,VEGFA,TNF,JUN,etc.GO biological process enrichment analysis screened the first 20 biological processes,including the response to toxic substances,the response to inorganic substances,the response to lipopolysaccharide,the response to injury,the metabolism of reactive oxygen species,and the apoptosis signaling pathway;the KEGG pathway enrichment analysis screened for 12 pathways associated with gastric cancer,including cancer pathway,IL-17,PI3 K-Akt signaling pathway,etc.Conclusion Atractylodes-Astragalus drug pair may mediate cancer pathways,IL-17,PI3 K-Akt and other signaling pathways,tumor cell response to toxic substances,inorganic substances,lipopolysaccharides,and regulation of tumor cell damage,oxygen metabolism,biological processes such as proliferation and apoptosis then play a role in the treatment of gastric cancer.
