摘要
目的探究欣胃颗粒对胃癌前病变(PLGC)模型大鼠胃黏膜中信号传导转录激活因子-3(STAT3)、存活素(Survivin)表达的影响。方法采用N-甲基-N′-硝基-N′-亚硝基胍(MNNG)为主的联合造模法建立PLGC模型大鼠120只,按随机数字表法分成空白组、模型组、欣胃颗粒组和维酶素组,欣胃颗粒组分为低、中、高剂量组,共6组,每组20只大鼠,分别给予模型大鼠生理盐水、欣胃颗粒低、中、高剂量、维酶素连续治疗,12周后,观察大鼠胃黏膜病理变化,及胃黏膜STAT3、Survivin基因的表达情况。结果与空白组比较,模型组、欣胃颗粒组、维酶素组STAT3 mRNA、Survivin mRNA表达水平相对均增高(P<0.05);与模型组比较,欣胃颗粒组、维酶素组STAT3 mRNA、Survivin mRNA表达水平相对均偏低(P<0.05);与维酶素组比较,欣胃颗粒组STAT3 mRNA、Survivin mRNA表达水平相对均偏低(P<0.05);与欣胃颗粒中剂量组比较,欣胃颗粒高、低剂量组表达水平相对均偏高(P<0.05),各组间差异具有统计学意义(P<0.01)。结论欣胃颗粒可有效改善胃癌前病变模型大鼠胃黏膜的病理学改变,且中计量效果相对最好,其机制可能与其调节STAT3/Survivin信号途径,下调模型大鼠胃黏膜的STAT3、Survivin基因表达水平相关。
Objective To explore the effect of Xinwei Granule(欣胃颗粒)on the expressions of signal transduction transcription activator-3(STAT3)and Survivin in gastric mucosa of gastric precancerous lesions(PLGC)model rats.Methods The N-methyl-N′-nitro-N′-nitrosoguanidine-based joint modeling method was used to establish 120 PLGC model rats,and they were divided into blank group and model group,Xinwei Granule low,medium and high dose groups and vinmycin group according to random number table method,20 rats in each group.The model group,Xinwei Granule low,medium and high dose groups and vinmycin group were given normal saline,Xinwei Granule low,medium and high dose and virmycin,respectively.After 12 weeks of continuous treatment,the pathological changes of the gastric mucosa and the expressions of STAT3 and Survivin genes in the gastric mucosa were observed.Results Compared with those of the blank group,the expression levels of STAT3 mRNA and Survivin mRNA in the model group,the Xinwei Granule groups and the vinmycin group were relatively higher(P<0.05).Compared with those of the model group,the expression levels of STAT3 mRNA and Survivin mRNA in the Xinwei Granule groups and the vinmycin group were relatively low(P<0.05).Compared with those of the vinmycin group,the expression levels of STAT3 mRNA and Survivin mRNA in the Xinwei Granule groups were relatively low(P<0.05).Compared with those of the Xinwei Granule medium dose group,the expression levels of Xinwei Granule high and low dose groups were relatively high(P<0.05),and the difference among the groups was statistically significant(P<0.01).Conclusion Xinwei Granule can effectively improve the pathological changes of gastric mucosa in rats with gastric precancerous lesions,and the effect of Xinwei Granule medium dose group is relatively best.Its mechanism may be related to the regulation of STAT3/Survivin signaling pathway and down-regulation of STAT3 and Survivin gene expression level in gastric mucosa of model rats.
