摘要
目的:优选肉桂挥发油的提取及β-环糊精包合工艺。方法:在对单因素实验结果分析的基础上,采取正交试验法,以挥发油提取率为指标,考察浸泡时间、加水量、提取时间3个因素,优选出挥发油最佳提取工艺;以包合物含油率、包合率及包合物得率为指标,考察β-CD和水比例、挥发油和β-CD的比例、包合温度、包合时间4个因素,优选挥发油β-环糊精最佳包合工艺;并用显微鉴别法、薄层色谱法对包合物进行验证及质量评价。结果:肉桂挥发油的最佳提取工艺为肉桂加8倍量的水浸泡1 h,提取5 h;最佳β-环糊精包合工艺为β-CD和水比例为1∶10,肉桂挥发油与β-CD的比例为1∶6,在包合温度为50℃下包合1.5 h。2项挥发油包合物验证实验证明挥发油包合物已经形成。结论:肉桂挥发油提取及β-环糊精包合工艺效果佳、重复性好。
Objective:The extraction of volatile oil from cinnamomum and the Pac-kage.and process ofβ-Cyclodextrin.Methods:Based on the analysis of the results of single factor experiments,the optimum extraction process of volatile oil was selected by using orthogonal test method and taking the content of volatile oil as the index,and investigating three factors:soaking time,adding water quantity and extracting time.Taking the oil content,inclusion rate and inclusion compound yield as indicators,the 4 factors,including theβ-Cyclodextrin to water,the ratio of volatile oil andβ-Cyclodextrin,inclusion temperature and inclusion time,were investigated,and the best inclusion process of volatile oil was determined.The inclusion compound was verified by microscopic identification and TLC and its quality was evaluated.Results:The optimum extraction technology of the essential oil of cinnamon was that the cinnamon was soaked in 8 times of water for 1 hour and extracted for 5 hours.The optimumβ-Cyclodextrin inclusion process wasβ-Cyclodextrin and water at 1∶10,and the proportion of cinnamon volatile oil andβ-Cyclodextrin was 1∶6,and 1.5 h was encapsulated at the inclusion temperature of 50℃.The two essential oil inclusion complexes were proved to be formed by the validation experiments.Conclusion:The extraction of volatile oil from Cinnamomum cinnamomi and the inclusion process ofβ-Cyclodextrin are good and reproducible.
