摘要
目的探讨磨盘草乙酸乙酯部位提取物对肝硬化腹水的影响及利尿作用。方法采用四氯化碳(CCl_(4))-橄榄油致大鼠肝腹水模型观察磨盘草乙酸乙酯部位提取物对肝硬化腹水的影响,测定血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、白蛋白(ALB)、醛固酮(ALD)水平及钾离子(K^(+))浓度并做病理检查;通过大鼠水负荷实验探讨利尿作用以及采用高效液相色谱法(HPLC)对磨盘草乙酸乙酯部位含药血清和空白血清,含药尿液和空白尿液进行初步成分分析。结果磨盘草乙酸乙酯部位可降低ALT、AST及ALD水平,可引起大鼠尿量增加且与药量呈现量效关系,发现并标定了12个入血成分及7个代谢产物。结论磨盘草乙酸乙酯部位具有消退腹水、改善肝功能及利尿作用,入血成分初步判定来源于磨盘草乙酸乙酯部位原药材或是其次级代谢产物。
Objective To discuss the effect of ethyl acetate extract of Abutilon indicum(L.)Sweet on cirrhosis ascites andiuretic effect.Methods The effect of the extracts of model on cirrhotic ascites caused by CCl_(4)-olive oil was observed.Serum AST,ALT,ALB,K^(+)and ALD levels were measured and pathological examination was performed.The diuretic effect was studied by water load experiment in rats and the preliminary composition analysis of drug serum and urine,blank serum and blank urine in ethyl acetate extract of Abutilon indicum(L.)Sweet was conducted by HPLC.Results The ethyl acetate extract of Abutilon indicum(L.)Sweet could reduce ALT,AST and ALD,and increase urine output in rats,which was in dose-effect relationship with the dose.Twelve components andseven metabolites were found and emarcated.Conclusion The ethyl acetate extract of Abutilon indicum(L.)Sweet has the effect of subsiding ascites,improving liver function and diuresis.The composition into blood is preliminarily determined to be from ethyl acetate extract of Abutilon indicum(L.)Sweet or the secondary metabolite.
