摘要
探究火炭母胶囊(HTM)对四氯化碳(CCl_(4))所致肝纤维化大鼠的影响及作用机制。方法:将72只雄性SD大鼠随机分为正常组、模型组、水飞蓟素组、HTM低剂量组(HTM-L,176 mg/kg)、HTM中剂量组(HTM-M,353 mg/kg)和HTM高剂量组(HTM-H,706 mg/kg),每组12只。除正常组大鼠外,其余大鼠按3 mL/kg腹部皮下注射40%CCl_(4)橄榄油液,每周2次,持续8周。造模开始同时药物干预,正常组大鼠按10 mL/kg灌胃给予0.5%羧甲基纤维素钠(CMC-Na),其余各组大鼠灌胃给予相应浓度药液,每天1次,持续8周。末次药后1h,处死大鼠,收集血液和肝组织。检测血清中丙氨酸氨基转移酶(ALT),天冬氨酸氨基转移酶(AST),白蛋白(ALB)活性及透明质酸(HA),层粘连蛋白(LN),Ⅲ型前胶原(PC-Ⅲ),Ⅳ型胶原(Col-Ⅳ),羟脯氨酸(HYP),白细胞介素IL-1β、IL-6、IL-10,肿瘤坏死因子-α,丙二醛(MDA),超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)的含量;用苏木素-伊红(HE)染色法观察各组大鼠肝组织病理学变化。结果:与模型组相比,HTM各剂量组大鼠纤维化程度减轻,体重增加,并降低肝纤维化指标ALT、AST水平(P<0.05);血清中HA、LN、PC-Ⅲ、Col-Ⅳ、HYP和MDA水平均降低(P<0.05),SOD和GSH水平均升高;HTM各剂量组大鼠血清IL-1β,中高剂量组IL-6及高剂量组TNF-α水平显著降低(P<0.05)。火炭母胶囊对四氯化碳诱导的大鼠肝纤维化有良好的改善作用。
Objective:To observe the effects and possible mechanisms of Polygonum chinense L.capsules(HTM)on hepatic fibrosis rats induced by carbon tetrachloride(CCl_(4)).Methods:72 male SD rats were randomly divided into control group,model group,silymarin group,low-dose HTM group(HTM-L,176 mg/kg),medium-dose HTM group(HTM-M,353 mg/kg),and high-dose HTM group(HTM-H,706 mg/kg)with 12 in each group.Except for the control group,the other rats were injected with 40%CCl_(4)-olive oil solution subcutaneously in the abdomen at 3 mL/kg,twice a week,total 8 weeks.Model making and drug intervention were carried out at the same time,rats in control group were given 0.5%CMC-Na at 10 mL/kg and other rats were given the corresponding concentration of medicine,once a day,total 8 weeks.After the last administration,the mice were sacrificed,and serum and liver tissue were collected.Serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),albumin(ALB),hyaluronic acid(HA),laminin(LN),typeⅢcollagen(PC-Ⅲ),typeⅣcollagen(Col-Ⅳ),hydroxyproline(HYP),interleukin IL-1β,IL-6,IL-10,tumor necrosis factor-α(TNF-α),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione(GSH)levels were measured in each group.The pathological changes were observed by HE staining.Results:Compared with the model group,HTM significantly increased the body mass of rats,alleviated the degree of liver damage,and reduced serum ALT and AST levels(P<0.05).The levels of HA,LN,PC-Ⅲ,Col-Ⅳ,HYP and MDA in serum was significantly decreased(P<0.05),SOD and GSH was significantly increased(P<0.05).The levels of serum IL-1βin each dose group of HTM,IL-6 in HTM-M and HTM-H,TNF-αin HTM-H were significantly decreased(P<0.05).Conclusion:HTM has markedly effect on improving liver fibrosis induced by CC14.
