摘要
目的:利用性状特征与内在质量的相关分析,探讨知母药材等级的划分依据,建立知母商品等级标准。方法:采用色差仪测定市售31批知母药材粉末颜色,并量化知母药材根长度、根中径等性状指标;采用HPLC法测定知母药材中芒果苷及知母皂苷BⅡ含量,同时测定知母水溶性浸出物、稀乙醇浸出物的含量;并对知母药材的外观性状及内在质量指标进行相关性、主成分及正交偏最小二乘判别分析,初步筛选出影响知母等级的主要指标。结果:芒果苷、水溶性浸出物、稀乙醇浸出物与根长度、根中径、a^(*)、b^(*)呈极显著正相关(P<0.01),且与L;呈极显著负相关(P<0.01),知母皂苷BⅡ相关系数较小,所筛选出的8个指标可将知母药材划分为4个等级,其中稀乙醇浸出物、L^(*)、b^(*)为影响等级划分的重要指标。结论:该研究所建立的质量等级标准与传统外观性状等级划分的结果基本一致,可以作为知母药材等级划分的依据。
Objective:To explore the grading basis of Anemarrhenae Rhizoma by correlation analysis between characters and internal quality,and to establish the grading standard of Anemarrhenae Rhizoma.Methods:The powder color of 31 batches of Anemarrhenae Rhizoma on the market was determined by colorimeter,and the root length,root diameter and other traits were quantified;HPLC method was used to determine the contents of mangiferin and timosaponin BⅡ,and the contents of water-soluble extract and dilute ethanol extract of Anemarrhenae Rhizoma were determined.Correlation analysis,principal component analysis and orthogonal partial least squares discriminant analysis were carried out on the appearance characters and internal quality indexes of Anemarrhenae Rhizoma,and the main indexes affecting the grade of Anemarrhenae Rhizoma were screened preliminarily.Results:Mangiferin,water-soluble extract,dilute ethanol extract had significant positive correlations with root length,root diameter,a^(*),b^(*),and negative correlations with L^(*)(P<0.01),the correlation coefficient of timosaponin BⅡ was small,the screened 8 indicators could be divided into four grades of Anemarrhenae Rhizoma,among which dilute ethanol extract,L^(*),b^(*)were the important indexes affecting the grade classification.Conclusion:The quality grade standard established in this study is basically consistent with the results of the traditional appearance character grade classification,which can be used as the basis for the grade classification of Anemarrhenae Rhizoma.
