摘要
本文通过网络药理学筛选补虚消渴合剂联合辅酶Q10对2型糖尿病的作用通路,通过2型糖尿病模型明确其作用效果。通过数据库筛选出疾病与药物相关基因,对二者共同靶点进行筛查。利用Cytoscape软件建立“药物-有效成分-靶点”及蛋白-蛋白相互作用的分子机制、GO功能分析和KEGG通路分析、AutoDockTools-1.5.7将核心靶点与对应有效成分进行分子对接。试验测定大鼠给药前后体重变化、空腹血糖(FBG)变化、试剂盒法测定血清葡萄糖(GLU)、糖化血清蛋白(GSP)、胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、胰岛素敏感指数(ISI)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量。网络药理学结果显示,补虚消渴合剂联合辅酶Q10与T2DM的共有靶点包括INS、AKT1、TNF、IL6、VEGFA等,这些靶点在糖尿病治疗中发挥关键作用,可通过降低胰岛素抵抗来调控糖尿病患者糖脂代谢。分子对接结果表明,核心靶点与相应的有效成分有结合活性。动物试验结果表明,补虚消渴合剂联合辅酶Q10明显降低糖尿病大鼠的血糖、血脂指标,改善胰岛素抵抗,调节胰岛素水平。补虚消渴合剂联合辅酶Q10可以通过多种成分、多个靶点、多条通路治疗T2DM,网络药理学与模型试验互相验证,为深入研究补虚消渴合剂联合辅酶Q10治疗2型糖尿病糖脂代谢紊乱提供了依据。
To screen the action pathway of Buxu Xiaoke mixture combined with coenzyme Q10 on type 2 diabetes mellitus through network pharmacology,and to clarify its effect through type 2 diabetes model.Screening the genes related to diseases and drugs with the help of database.Screening the common targets of the two.The molecular mechanism of"drug-active ingredient-target"and protein-protein interaction,and the pathway analysis of GO and KEGG were established by Cytoscape software.Using AutoDockTools-1.5.7 to make molecular docking between the core target and the corresponding effective components.The changes of body weight,measurement of changes in fasting blood glucose(FBG),serum glucose(GLU),glycated serum protein(GSP),insulin(FINS)content,insulin resistance index(HOMAIR),insulin sensitivity index(ISI),total cholesterol(TC),triglyceride(TG)and low density fat were determined by kit method before and after administration protein cholesterol(LDL-C)content.The results of network pharmacology showed that the common targets of Buxu Xiaoke mixture combined with coenzyme Q10 and T2DM include INS,AKT1,TNF,IL-6 and VEGFA,which play a key role in the treatment of diabetes and regulates glucose and lipid metabolismin diabetic patients by reducing insulin resistance.The results of molecular docking sho-wed that the core target has binding activity with the corresponding effective components.The results of animal experiments showed that Buxu Xiaoke mixture combined with coenzyme Q10 significantly reduced the blood sugar and lipid indexes of diabetic rats,and improved insulin resistance by regulating insulin levels.Buxu Xiaoke mixture combined with coenzyme Q10 can treat T2DM through multi-component,multi-target and multi-pathway effects,and the network pharmacology and experimental results are mutually verified.This study provides a basis for further study of Buxu Xiaoke mixture combined with coenzyme Q10 in the treatment of type 2 diabetes mellitus with glucose and lipid metabolism disorders.
