摘要
目的探究复方百部止咳糖浆对百日咳小鼠呼吸道感染模型免疫功能、Toll样受体4/核因子-κB(TLR4/NF-κB)信号通路的影响。方法使用TohamaⅠ株感染7 d建立百日咳小鼠呼吸道感染模型,将造模成功小鼠随机分为模型组、阳性组(40 mg/kg罗红霉素混悬液)和复方百部止咳糖浆低、中、高剂量组(0.715、1.43、2.86 g/kg),每组12只;另取12只正常小鼠作为对照组。给药3 d后,观察小鼠状态并进行菌落计数,HE染色观察肺组织形态,ELISA法检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6水平,流式细胞仪检测腹主动脉血中CD4^(+)、CD8^(+)T细胞比例,Western blot法检测肺组织TLR4、NF-κB蛋白表达。结果与对照组比较,模型组小鼠状态较差,肺组织病理损伤明显,菌落数量,肺组织损伤评分,血清TNF-α、IL-1β、IL-6水平,腹主动脉血CD8^(+)T细胞比例,肺组织TLR4、NF-κB蛋白表达升高(P<0.05),腹主动脉血CD4^(+)T细胞比例降低(P<0.05);与模型组比较,复方百部止咳糖浆各剂量组上述各项指标均得以逆转(P<0.05),并呈剂量依赖性;阳性组效果优于复方百部止咳糖浆低、中剂量组,但不及高剂量组。结论复方百部止咳糖浆可缓解百日咳杆菌呼吸道感染小鼠炎症因子水平并参与免疫调节,其作用可能与抑制TLR4/NF-κB信号通路有关。
AIM To explore the effects of Compound Baibu Zhike Syrup on immune function and Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)signaling pathway in the mouse model of pertussis induced by respiratory tract infection.METHODS The successful mouse models of pertussis established by 7 days exposure to TohamaⅠstrain were randomly divided into the model group,the positive group(40 mg/kg roxithromycin suspension)and the low,medium and high dose groups of Compound Baibu Zhike Syrup(0.715,1.43 and 2.86 g/kg),with 12 mice in each group,in contrast to another 12 normal mice of the control group.After 3 days of corresponding administration,the mice had their general conditions observed and the colony count made;their pulmonary morphology observed by HE staining;their levels of serum tumor necrosis factor-α(TNF-α),interleukin(IL)-1βand IL-6 detected by ELISA;their ratios of CD4^(+)and CD8^(+)T cells in abdominal aortic blood detected by flow cytometry;and their protein expressions of TLR4 and NF-κB in pulmonary tissue detected by Western blot.RESULTS Compared with the control group,the model group was found with poor general conditions;obvious pulmonary pathological injury;increased colony number,pulmonary injury score,serum TNF-α,IL-1βand IL-6 levels,ratio of CD8+T cells in abdominal aorta blood,protein expressions of TLR4 and NF-κB in pulmonary tissue(P<0.05);and decreased the ratio of CD4^(+)T cells in abdominal aorta blood(P<0.05).Compared with the model group,Compound Baibu Zhike Syrup groups shared obviously different conditions in a dose-dependent manner in terms of the all reversed aforementioned indices levels(P<0.05).However,the positive group demonstrating a better effect than the low and middle dose Compound Baibu Zhike Syrup groups was inferior to the high dose group.CONCLUSION Being an immunoregulator via TLR4/NF-κB signaling pathway,Compound Baibu Zhike Syrup can alleviate the levels of inflammatory factors in mice infected with Bordetella pertussis through respiratory tract.
