川芎生物碱对大鼠心肌纤维化的影响 被引量:5

Effects of Chuanxiong Alkaloid on Myocardial Fibrosis in Rats

摘要 目的探讨川芎生物碱对异丙肾上腺素(Iso)诱导大鼠心肌纤维化(MF)后心肌组织中羟脯氨酸(Hyp)含量、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及血清中乳酸脱氢酶(LDH)和肌酸激酶(CK)活性的影响.方法将50只大鼠随机分成对照组、模型组、川芎生物碱低中高剂量治疗组共5组,每组10只.应用Iso建立大鼠MF病理损伤模型,应用羟脯氨酸试剂盒检测心肌组织中羟脯氨酸的含量,黄嘌呤氧化酶法测定血清中SOD活性,硫代巴比妥酸显色法测定MDA含量,半自动生化分析仪测定LDH、CK活性.结果模型组与对照组比较,心肌组织中羟脯氨酸的含量升高,血清中的SOD活性降低,MDA含量增加(P<0.05),LDH和CK活性升高(P<0.01);川芎生物碱低中高剂量治疗组与模型组比较,血清中的SOD活性均升高,MDA含量均降低(P<0.05),LDH和CK活性均显著降低(P<0.01),心肌组织中Hyp含量降低(P<0.05或P<0.01).结论川芎生物碱可抑制大鼠MF,并对缺血的心肌具有保护性作用. Objective To investigate the effects of Chuanxiong alkaloid on the contents of hydroxyproline(Hyp),superoxide dismutase(SOD)and malondialdehyde(MDA)in myocardium and the activities of lactate dehydrogenase(LDH)and creatine kinase(CK)in serum after isoproterenol-induced myocardial fibrosis(MF)in rats. Method Fifty rats were randomly divided into 5 groups with 10 rats in each group,control group,model group,Chuanxiong alkaloid low,medium and high dose treatment group.The pathological damage model of MF was established by Iso.Hydroxyproline kit was used to detect the content of hydroxyproline in cardiac tissue.The activity of SOD in serum was determined by Xanthine oxidase method.The content of MDA was determined by thiobarbituric acid colorimetry.Activities of LDH and CK were measured by semi-automatic biochemical analyzer. Results Compared with control group,the content of hydroxyproline in myocardial tissue in model group incr-eased,the activity of SOD decreased,the content of MDA increased(P<0.05),the activities of LDH and CK increased(P<0.01).Compared with model group,SOD activity in serum increased,MDA content decreased(P<0.05),LDH and CK activity were significantly decreased(P<0.01),and Hyp content in myocardial tissue decreased in Chuanxiong alkaloid low-medium-high-dose treatment group. Conclusion Chuanxiong alkaloid can inhibit MF in rats and has a protective effect on ischemic myocardium.
