不同致病性的麦根腐平脐蠕孢菌代谢组学分析 被引量:1

摘要 为明确引起大麦叶斑病的不同麦根腐平脐蠕孢菌(Bipolaris sorokiniana)的致病性差异,通过大麦叶斑病苗期侵染型分级标准测定弱致病性麦根腐平脐蠕孢菌ND-85和强致病性麦根腐平脐蠕孢菌ND-87的致病性,进一步采用非靶向代谢组学与多元统计分析方法,比较了2种不同麦根腐平脐蠕孢菌菌株的代谢差异。主成分分析(PCA)结果显示,菌株ND-85和ND-87间分散明显,通过正交偏最小二乘法判别分析(OPLS-DA)共筛选出726种重叠的差异代谢产物,其中,有458种差异代谢产物下调,268种差异代谢产物上调;下调代谢产物主要包括有机酸、酯类物质等,上调差异代谢产物主要包括氨基酸、有机酸、硒等。菌株ND-85与ND-87通过KEGG注释得到的差异代谢物有66条代谢通路,其中,甘氨酸、丝氨酸和苏氨酸生物合成富集程度最为显著,ABC转运蛋白合成途径的富集程度最高。推测ABC转运蛋白合成、耐万古霉素合成、氨酰-tRNA生物合成、甘氨酸、丝氨酸和苏氨酸的生物合成、精氨酸和脯氨酸的代谢、赖氨酸降解途径与菌株的致病力差异有关。 To clarify the pathogenicity difference of different Bipolaris sorokiniana causing barley spot blotch disease,we verified the pathogenicity of ND-85 with the weak pathogenicity and ND-87 with the strong pathogenicity through the classification standard of infection type of barley spot blotch disease at barley seedling stage.In addition,non-targeted metabonomics and multivariate statistical analysis were used to compare the metabolic differences of two different Bipolaris sorokiniana isolates.Principal component analysis(PCA)results showed significant dispersion between the ND-85 and ND-87 strains.A total of 726 types of overlapping differential metabolites were screened by orthogonal partial least squares discriminant analysis(OPLS-DA),of which 458 types of metabolites were down-regulated,and 268 types of metabolites were up-regulated.The down-regulated metabolites mainly included organic acids and esters,and up-regulated metabolites mainly included amino acids,organic acids and selenium,etc.There were 66 types of metabolic pathways for the differential metabolites of ND-85 and ND-87 by KEGG annotation,among which the enrichment degree of glycine,serine and threonine biosynthesis was the most significant,and the enrichment degree of ABC transporter biosynthesis was the highest.It was speculated that the pathogenicity differences were related to the ABC transporter synthesis,vancomycin resistance,aminoacyl-tRNA-biosynthesis,glycine,serine and threonine biosynthesis,arginine and proline metabolism,and lysine degradation pathway.
