大熊猫源枝孢样枝孢霉对实验小鼠的血常规、生化及致病性影响的研究 被引量:1

The effects of blood routine,biochemistry and pathogenicity of Cladosporium cladosporioides isolated from giant panda in mice

摘要 为研究枝孢样枝孢霉大熊猫体表分离株对小鼠血液常规和生化指标的影响及其致病性,本研究将24只健康昆明小鼠,随机分为实验组(A)、免疫抑制组(B)和对照组(C),A组和B组小鼠经腹部皮下注射接种枝孢样枝孢霉菌(1.5×10^(6) cfu/mL)悬液0.2 mL,C组注射等量生理盐水。接种后第14 d和第28 d分别迫杀4只小鼠进行血常规、生化、病理组织学检查和真菌培养。结果显示:与对照组相比,A组和B组小鼠接种部皮肤及浅表组织出现结节和色素沉着,白细胞数、淋巴细胞数极显著降低(p<0.01),单核细胞数显著升高(0.01<p<0.05),A组小鼠中性粒细胞数显著降低(0.01<p<0.05);A组和B组小鼠总胆红素显著降低(0.01<p<0.05)。对测定的其余肝肾功能和血常规指标与对照组相比差异均不显著;病理学检查结果显示小鼠接种部位的白细胞、单核细胞和巨噬细胞等数量均增多,且发现大量的真菌孢子和菌丝;A组和B组小鼠皮肤组织真菌培养结果为枝孢样枝孢霉。综上表明,枝孢样枝孢霉感染小鼠后导致其局部皮肤及浅表组织白细胞、单核细胞和淋巴细胞聚集。本研究首次发现枝孢样枝孢霉感染小鼠后引起白细胞数、淋巴细胞数极显著降低,对其免疫相关细胞数有较大影响,可导致小鼠免疫功能下降,且对小鼠有较强致病性。本研究为科学防治大熊猫枝孢样枝孢霉引起的疾病提供科学依据。 To study the effect of Cladosporium cladosporioides isolated from the body surface of giant panda on blood routine and biochemical indicators in mice and its pathogenicity.Twenty-four healthy Kunming mice were randomly divided into experimental group(A),immunosuppressive group(B)and control group(C).Mice in groups A and B were all inoculated with 0.2mL of fungal spores suspension(1.5×106cfu/mL)by abdominal subcutaneous injection,and mice in group C were injected with the same amount of normal saline.Four mice were sacrificed on the 14th and 28th days after the inoculation for blood routine,biochemical and histopathological examination and reverse culture,respectively.In comparison with the control group,nodules and pigmentation appeared in the skin and superficial tissues of the inoculated area of mice in groups A and B;white blood cell count(WBC)and lymphocytes count were extremely significant reduced(p<0.01),monocytes were significantly reduced(0.01<p<0.05),and neutrophils in group A were significantly decreased(0.01<p<0.05);the total bilirubin(TBIL)of mice in groups A and B was significantly decreased(0.01<p<0.05).There was no significant difference in other liver and kidney functions and blood routine indexes.The results of pathological examination showed that there were more leukocytes,monocytes and macrophages at the inoculation site of mice,and a large number of fungal spores and hyphae were found.The results of fungus culture in the skin tissues of mice in groups A and B were C.cladosporium.Therefore,after inoculation of C.cladosporium in mice,leukocyte,monocyte and lymphocyte gathered in local skin and superficial tissue of mice.In summary,it was found for the first time that C.cladosporium infection in mice caused a very significant decrease in the number of white blood cells and lymphocytes,which had a great influence on the immune related cells,and could lead to a decline in the immune function of experimental mice,and had a strong pathogenicity to mice.This study provides scientific basis for scientific prevention and control of diseases caused by white blood cells in giant pandas.
