Establishment of animal models of premature ovarian failure

摘要 卵巢早衰(POF)常常导致生育力下降及低雌激素状态,严重影响女性的身心健康及生活质量。POF的发病机制尚不明确,通过动物模型探究其发病机制,寻找新的治疗途径具有重要的临床意义。本文按照遗传、自身免疫、医源性和代谢等POF的病因分类,阐述动物模型建立的具体方法。包括构建与卵泡发育相关的基因敲除鼠,皮下注射透明带抗原3(ZP3)、胸腺切除和自体卵巢主动免疫,放化疗使用X射线、γ射线及使用顺铂、环磷酰胺、表柔比星等化疗药物,及使用生殖毒性药物雷公藤制剂,皮下注射D-半乳糖构建半乳糖血症相关的POF。旨为POF的基础研究提供参考依据。 Premature ovarian failure(POF)leads to fertility decrease and low estrogen level,which seriously affects women's physical and mental health and quality of life.The pathogenesis of POF is still unclear.It is of great clinical significance to explore its pathogenesis and new therapeutic approaches through animal models.According to the classification of the etiology of POF,this article described the specific methods of establishing animal models from four aspects:heredity,autoimmunity,iatrogenic and metabolic factors.In terms of genetics,there are knockout mice related to follicular development.In immune aspects,methods are subcutaneous injection of zona pellucida antigen 3(ZP3),thymectomy,and autologous ovarian active immunity.Iatrogenic methods included radiotherapy(X-rays orγ-rays),chemotherapy(cisplatin,cyclophosphamide or epirubicin)and reproductive toxic drugs(tripterygium preparations).Animal models of POF relevant to galactosemia were constructed by subcutaneous injection of D-galactose.This article aims to provide reference for the basic research of POF.
