两古方在改善POI大鼠卵巢微环境作用机制的差异初探 被引量:3

Preliminary Study of the Difference Between Ancient Sides in Improving the Mechanism of Ovarian Environment in POI Rats

摘要 目的本文旨在探究毓麟珠与养精种玉汤在改善卵巢微环境进而改善早发性卵巢功能不全(Premature ovarian insufficiency,POI)的作用机制差异。方法选择SPF级雌性大鼠24只,分为4组,每组6只,除空白组外,其余各组通过腹腔注射去氧乙烯基环己烯(VCD)构造POI模型,造模成功后,空白组与模型组给予生理盐水灌胃,毓麟珠组与养精种玉汤组分别给与相应中药灌胃6周。通过HE染色确立模型构造成功;通过小动物多普勒超声系统检测卵巢血流及形态;通过ELISA检测血清激素水平;通过氧化应激因子超氧化物歧化酶(SuperoxideDismutase,SOD)、丙二醛(Malondialdehyde,MDA)、谷胱甘肽过氧化物酶(Glutathione Peroxidase,GSH-Px)检测卵巢氧化应激水平。结果HE染色结果显示,模型组大鼠卵泡数减少,透明带损伤,卵母细胞形态畸形,造模成功;超声数据显示与模型组相比,毓麟珠组卵巢面积明显增加(P<0.05),毓麟珠组与养精种玉汤组血流动力指数(PI)与阻力指数(RI)值明显降低,养精种玉汤组降低效果更明显;血清激素结果显示,与空白组相比,E2、AMH明显降低,FSH明显升高(P<0.05),给药后E2、AMH明显升高,FSH明显降低(P<0.05),毓麟珠改善效果更明显;氧化应激结果显示,模型组SOD、GSH-PX明显降低,MDA明显升高(P<0.05),给药6周后,养精种玉汤组SOD明显升高,MDA明显降低(P<0.05);毓麟珠组GSH-PX明显升高,MDA明显降低(P<0.05),养精种玉汤组改善更明显。结论毓麟珠组改善激素水平以及卵巢状态效果更明显,养精种玉汤组在改善卵巢血流以及氧化应激效果更好,说明中医同病异治理论有其充分的科学依据,为不同证型药物防治POI的机制探索提供思路。 Objective To explore the mechanism differences between YuLinzhu and Yangjing-Zhongyu decoction in improving ovarian microenvironment and then improving premature ovarian insufficiency(POI).Methods 24 female rats were divided into four groups.Except the blank group,each group constructed the POI model by peritoneal deethylene cyclohexene(VCD).After the mold creation,the blank group and the model group gave normal saline stomach,the YuLinzhu group and the Yangjing-Zhongyu decoction group gave correspondent drugs for 6 weeks.HE staining was used as a criterion for successful model construction;observation of ovarian blood flow and morphology by small animal Doppler ultrasound system;serum hormone levels by ELISA;and Ovarian oxidative stress levels were measured by detecting oxidative stress factor superoxide dismutase(SOD),maldialaldehyde(MDA)and glutathione peroxidase(GSH-Px).Results The HE staining results show that,the number of follicles in the model group of rat decreased,Clear band damage,oocyte morphological malformations,successful model making;Ultrasonic data showed that the ovarian area was significantly increased in Yulinzhu group compared with the model group(P<0.05),the hemodynamic index(PI)and resistance index(RI)values in Yulinzhu group were significantly reduced,and the reduction effect of the Yangjing-Zhongyu decoction group was more obvious;Serum hormone results showed that,compared to the blank groups,E2、AMH decreased significantly(P<0.05),FSH rises significantly(P<0.05),E2、AMH increased significantly after administration,FSH is significantly reduced(P<0.05),Yulinzhu group improvement effect is more obvious;Oxidative stress results show that,significant decrease in model group SOD、GSH-PX,MDA rises significantly(P<0.05),Six weeks after the administration,SOD is significantly increased in the Yangjing-Zhongyu decoction group,MDA decreased significantly(P<0.05);the GSH-PX of YuLinzhu group has increased significantly,the MDA is significantly reduced(P<0.05),Yangjing-Zhongyu decoction group has improved more significantly.Conclusion Yu Linzhu group to improve hormone level and ovarian status effect is more obvious,and YuLinzhu and Yangjing-Zhongyu decoction group is better in improving ovarian blood flow and oxidative stress,based on the overall concept of traditional Chinese medicine,provides some scientific basis in basic research,and provides some reference for syndrome differentiation in clinical treatment.
