摘要
畜禽肌内脂肪(IMF)含量影响肉质风味、口感、嫩度、色泽等指标。调节IMF含量改善肉质成为研究重点。通过添加不同浓度(0、0.15、0.30、0.45、0.60及0.75 mmol·L^(-1))缬氨酸,检测脂肪合成相关重要信号分子SREBP-1c蛋白表达与成熟情况,检验脂滴生成和细胞增殖情况,探究缬氨酸如何影响小鼠成肌细胞(C2C12)胞内脂肪沉积。结果表明,体外培养36 h后,0.45 mmol·L^(-1)缬氨酸处理C2C12,可显著促进SREBP-1c蛋白成熟、表达和细胞增殖,一定程度促进脂滴沉积;添加0.45 mmol·L^(-1)缬氨酸和75μmol·L^(-1)棕榈酸后C2C12细胞中SREBP-1c蛋白含量显著上调,进一步促进细胞内脂质沉积,C2C12细胞增殖未受抑制。综上,缬氨酸可通过正向调节SREBP-1c相关信号通路,参与C2C12细胞内脂肪沉积或转运。研究首次证实缬氨酸可促进小鼠C2C12细胞脂肪沉积及增殖,为进一步探究缬氨酸在脂肪沉积过程中作用提供新见解,也为缬氨酸提高畜禽肉质研究提供基础和依据。
The content of intramuscular fat(IMF)in livestock and poultry directly affects meat flavor,taste,tenderness,color and other indicators.In recent years,improving meat quality by adjusting IMF content has become the focus of scientific research.Objective of the current research was to explore the effect of valine treatment on C2C12 cells on intra-cellular fat deposition.After treating C2C12 with different concentrations of valine(0,0.15,0.30,0.45,0.60 and 0.75 mmol·L^(-1)),the expression and maturation of SREBP-1c(an important signaling molecule related to fat synthesis)were detected,the formation of lipid droplets and cell proliferation were also checked.The results showed that after 36 h of culture in vitro,0.45 mmol·L^(-1) valine treatment of myoblasts could significantly promote the maturation and expression of SREBP 1c protein,as well as cell proliferation,and promote lipid droplet deposition to a certain extent.When 0.45 mmol·L^(-1) valine and 75μmol·L^(-1) palmitic acid were added simultaneously,the content of SREBP-1c protein in C2C12 cells was significantly increased,and lipid deposition in C2C12 cells was further promoted.However,the proliferation of C2C12 cells was not inhibited.Taken together,valine might participate in fat deposition or transport in C2C12 cells through positive regulation of SREBP 1c-related signaling pathways.The current research confirmed for the first time that valine could promote fat deposition and proliferation of mouse C2C12 cells.It provided new insights for further exploring the role of valine in fat deposition,and experimental and theoretical basis for valine to improve the meat quality of livestock and poultry.
