摘要
目的:探究理中汤加味对大鼠复发性口腔溃疡(ROU)黏膜的修复作用及机制。方法:60只Wistar大鼠随机分为正常组、模型组、阳性对照组(醋酸泼尼松龙,1 mg/mL,20 mL/kg)、理中汤高(40 g/kg)、中(20 g/kg)、低剂量组(10 g/kg),每组10只。除正常组外其余5组建立ROU模型,建模8周后按分组灌胃给药(100 mL/100 g),连续20 d。然后计算大鼠口腔溃疡数量、持续和间隔时间、溃疡面积,HE染色观察口腔黏膜组织病变,ELISA法检测血清IL-1β、IL-6、TNF-α水平,Western blot法检测溃疡组织NF-κB炎症通路蛋白表达。结果:理中汤加味减轻了大鼠口腔黏膜损伤和炎症浸润,促进溃疡修复,降低IL-1β、IL-6、TNF-α水平和IKKα、p-IκBα、NF-κB p65蛋白表达(P<0.05),理中汤效果呈剂量依赖性(P<0.05),高剂量组作用强于阳性对照组(P<0.05)。结论:理中汤加味对ROU模型大鼠黏膜有修复作用,其机制可能与抑制NF-κB炎症通路有关。
Objective:To explore the repairing effect and mechanism of Lizhong Decoction on recurrent oral ulcer(ROU)in rats.Methods:60 Wistar rats were randomly divided into normal group,model group,positive control(predisone acetate,1 mg/mL,20 mL/kg)group,and Lizhong Decoction high(40 g/kg),medium(20 g/kg)and low(10 g/kg)dose groups(n=10).Except for normal group,ROU model was established in the other 5 groups.8 weeks after model establishment the drugs were given by gavage(100 mL/100 g)for 20 days.Then the number,duration,interval time and oral ulcer area in each group of the rats were calculated.HE staining was used to observe the pathological changes of oral mucosa.The serum levels of IL-1β,IL-6 and TNF-αwere detected by ELISA.Western blot method was used to detect the protein expression level of NF-κB inflammation pathway in ulcer tissues.Results:Lizhong decoction reduced oral mucosal damage and inflammation in the rats,promoted ulcer repair,and reduced IL-1β,IL-6,TNF-αlevels and IKKα,p-IκBα,and NF-κB p65 protein expression in a dose dependent mannet(P<0.05),40 g/kg of Lizhong decoction showed stronger effects than the positive control(P<0.05).Conclusion:Lizhong decoction has a repairing effect on the mucosa with ROU in rats,and its mechanism may be related to the inhibition of NF-κB inflammation pathway.
