摘要
目的:探讨益心泰颗粒对慢性心力衰竭心气虚兼血瘀水停证大鼠心肌AMPK蛋白表达的影响。方法:采用丙基硫氧嘧啶(PTU)混悬液灌胃及阿霉素(ADR)腹腔注射复制慢性心力衰竭心气虚兼血瘀水停证大鼠模型。将造模成功的大鼠随机分为模型组、益心泰高剂量组、益心泰中剂量组、益心泰低剂量组和曲美他嗪组;另设正常对照组。各药物组给予相应药物灌胃,模型组和正常对照组灌胃同等体积的蒸馏水,1次/d,共4周。实验结束后检测大鼠血清NT-proBNP、FFA及心肌Na^(+),K^(+)-ATP酶活性、Ca^(2^(+)),Mg^(2^(+))-ATP酶活性、AMPK蛋白表达。结果:与模型组比较,各药物组血清NT-proBNP、FFA显著降低(P<0.01),而心肌Na^(+),K^(+)-ATP酶、Ca^(2^(+)),Mg^(2^(+))-ATP酶活性、AMPK蛋白相对灰度值均显著升高(P<0.01)。与益心泰低剂量组比较,益心泰中、高剂量组血清NT-proBNP、FFA均明显降低(P<0.05),而心肌Na^(+),K^(+)ATP酶活性、Ca^(2^(+)),Mg^(2^(+))-ATP酶活性、AMPK蛋白相对灰度值均明显升高(P<0.05)。结论:益心泰颗粒可能通过上调心肌AMPK蛋白表达水平,降低血清NT-proBNP、FFA水平,增加ATP酶活性。
Objective:To investigate the effect of Yixintai Granules on the expression of AMPK protein in the myocardium of rats with chronic heart failure of heart Qi deficiency and blood stasis and water stagnation syndrome.Methods:Propylthiouracil(PTU)suspension was used to gavage and adriamycin(ADR)intraperitoneally injected to replicate a rat model of chronic heart failure of heart Qi deficiency and blood stasis and water stagnation syndrome.The successfully modeled rats were randomly divided into model group,Yixintai high-dose group,Yixintai medium-dose group,Yixintai low-dose group and trimetazidine group.Besides,a normal control group was also set up.Rats in each administration group were given corresponding drugs,and the model group and the normal control group were given the same volume of distilled water once a day for 4 weeks.After the experiment,the rat serum NT-proBNP,FFA,myocardial Na^(+)/K^(+)ATPase,Ca^(2^(+))/Mg^(2^(+))ATPase activity,and AMPK protein expression were detected.Results:Compared with the model group,the serum NT-proBNP and FFA of e ach administration group were significantly decreased(P<0.01),while the myocardial Na^(+)/K^(+)ATPase,Ca^(2^(+))/Mg^(2^(+))ATPase activity and the relative gray value of AMPK protein were significantly increased(P<0.01).Compared with Yixintai low-dose group,serum NT-proBNP and FFA of Yixintai middle and high-dose groups were significantly decreased(P<0.05),while myocardial Na^(+)/K^(+)ATPase,Ca^(2^(+))/Mg^(2^(+))ATPase activityand the relative gray value of AMPK protein were significantly increased(P<0.05).Conclusion:Yixintai Granules may increase the level of myocardia AMPK protein,decrease serum NT-proBNP and FFA levels,and increase ATPase activity.
