摘要
目的:研究沙棘黄酮对离体蛙心心肌收缩力的影响并探讨其可能机制。方法:用保险粉(Na2S2O4)造成H9C2心肌细胞缺氧损伤,分别用沙棘黄酮高、中、低浓度作用于H9C2细胞缺氧损伤模型,以细胞存活率为指标探究沙棘黄酮对心肌细胞的保护作用。随后采用八木氏法制备离体蛙心灌流标本,分别加入不同浓度沙棘黄酮,研究不同浓度下沙棘黄酮对蛙心肌收缩力、心率和心输出量的影响。通过给予乙酰胆碱、阿托品、肾上腺素及低钙、高钙和正常任氏液,探究沙棘黄酮影响蛙心收缩功能的作用及机制。结果:沙棘黄酮可明显提高心肌细胞的存活率,可使蛙心心肌收缩力、心输出量明显降低(P<0.01),心率减慢(P>0.05);M受体阻断剂阿托品不能拮抗沙棘黄酮抑制心肌收缩的作用,沙棘黄酮也不能完全拮抗肾上腺素增强心肌收缩力的作用;沙棘黄酮在低钙、高钙和正常任氏液中抑制离体蛙心的作用随细胞外钙离子浓度的升高而增强(P<0.01)。结论:沙棘黄酮对心肌细胞具有保护作用,对离体蛙心心肌收缩具有抑制作用,其作用机制可能与抑制细胞外钙离子内流有关。
Objective:To study the effect of seabuckthorn flavone on the cardiac functions of isolated frog heart and its possible mechanism.Methods:H9C2myocardial cells were treated with insurance powder(Na2S2O4)for hypoxia injury.The H9C2cell model was treated with high,medium and low concentrations of seabuckthorn flavonoids respectively.The protective effect of seabuckthorn flavonoids on myocardial cells was investigated by using cell survival rate as index.The frog heart-intubation method was adopted,the isolated frog hearts were perfusion with different doses of seabuckthorn flavone to study the effects on cardiac contractility,heart rate and cardiac output.The effects and mechanisms of seabuckthorn flavone on the contractile function of frog heart were studied by administration of acetylcholine,atropine,epinephrine,low calcium,high calcium and normal Ringer’s fluid.Results:Seabuckthorn flavone can obviously improve the survival rate of myocardial cells,significantly decrease cardiac contractility and cardiac output(P<0.01)and slow down heart rate(P>0.05).Atropine,an M receptor blocker,could not antagonize the effect of seabuckthorn flavone on myocardial contraction,seabuckthorn flavone could not completely antagonize the effect of adrenaline on myocardial contractility.The inhibitory effect on isolated frog heart increased with the increase of extracellular calcium concentration of the Ringer’s fluid(P<0.01).Conclusion:seabuckthorn flavone have protective effect on myocardial cells and inhibitory effect on myocardial contraction in isolated frog heart.The mechanism may be related to the inhibition of extracellular calcium influx.
