启膈散联合顺铂对与成纤维细胞共培养的食管癌EC9706细胞生长的影响 被引量:4

Effect of Qige Powder Combined with Cisplatin on the Growth of Esophageal Cancer Cells EC9706 Co-cultured with Fibroblasts

摘要 目的研究与成纤维细胞共培养条件下的食管癌细胞对顺铂敏感性变化及启膈散干预作用。方法建立食管癌细胞和成纤维细胞共培养方法,MTT法检测顺铂、启膈散、或两者联合应用对细胞生长的影响,流式细胞术检测细胞周期和凋亡。结果在顺铂相同浓度作用下,共培养条件下食管癌EC9706组OD值明显高于单独培养食管癌EC9706组(P<0.05);共培养条件下顺铂的IC50值明显高于单独培养食管癌EC9706组(P<0.05);不同浓度启膈散乙酸乙酯提取物对共培养条件下食管癌EC9706细胞有明显的抑制作用,呈浓度剂量依赖性,与顺铂组比较,联合用药组OD值明显减少,联合用药有协同作用趋势(P<0.05);与单独培养EC9706组比较,共培养条件组总凋亡率下降,S期比例增加(P<0.05);与共培养组比较,顺铂组明显使EC9706早期凋亡率增加,而启膈散组,联合用药组均使EC9706晚期凋亡率增加(P<0.05);顺铂组、启膈散组及联合用药组明显降低了EC9706细胞G0/G1期的比例,而增加了EC9706细胞S期的比例(P<0.05),其中以联合用药组作用最明显(P<0.05)。结论启膈散可抑制与成纤维细胞共培养条件下的食管癌细胞增殖、诱导凋亡,干预细胞周期,与顺铂联合应用具有一定增效作用。 Objective To study the sensitivity of esophageal cancer cells to cisplatin and the intervention of Qige Powder under the condition of esophageal cancer cells EC9706 being co-cultured with fibroblasts.Methods The esophageal cancer cells were co-cultured with the fibroblasts,and then the effect of cisplatin and Qige Powder alone or used together on the cell growth was detected by MTT method,and cell cycle and apoptosis were detected by flow cytometry.Results After treatment with the same concentration of cisplatin,the OD value in the co-culture group of esophageal cancer cells EC9706 together with the fibroblasts was higher than that in the single-culture group of EC9706(P<0.05).And the IC50 value of cisplatin in the co-culture group was significantly higher than that in the single-culture EC9706 group(P<0.05).Different concentrations of Qige Powder ethyl acetate extract had significant inhibitory effect on EC9706 cells co-cultured with the fibroblasts,and the effect was in dosedependent manner.Compared with the cisplatin group,the OD value of cisplatin combined with Qige Powder group was significantly reduced,and the combination of cisplatin and Qige Powder drug showed a synergistic effect(P<0.05).Compared with single-culture group of EC9706,the total apoptotic rate of co-culture group was decreased,and the proportion of EC9706 cells at S phase was increased(P<0.05).Under the condition of the coculture,cisplatin alone significantly increased the early apoptotic rate of EC9706,while Qige Powder alone and the combination of cisplatin and Qige Powder increased the late apoptotic rate of EC9706(P<0.05);Cisplatin alone,Qige Powder alone and the combination of Cisplatin and Qige Powder significantly reduced the proportion of EC9706 cells at G0/G1 phase,but increased the proportion of EC9706 cells at S phase(P<0.05),and the combination of the two drugs had the strongest effect(P<0.05).Conclusion Qige Powder can inhibit the proliferation and induce the apoptosis of esophageal cancer cells,and interfere with the cell cycle of the esophageal cancer cells co-cultured with the fibroblasts.The medication of Qige Powder combined with cisplatin has a synergistic effect.
