高效液相色谱法同时测定食品中安赛蜜、苯甲酸( 钠) 、山梨酸( 钾) 、脱氢乙酸( 钠) 和糖精钠 被引量:28

Simultaneous determination of acesulfame potassium,benzoic acid(sodium),sorbic acid(potassium),dehydroacetic acid(sodium)and saccharin sodium in food by high-performance liquid chromatography

摘要 目的研究一种快速、准确、简单的高效液相色谱法同时测定食品中安赛蜜、苯甲酸(钠)、山梨酸(钾)、脱氢乙酸(钠)和糖精钠等5种添加剂。方法根据样品类别选择不同的前处理方法,以甲醇∶乙酸铵(pH=6.8)=5∶95(体积比)作为流动相,利用二极管阵列检测器(DAD)检测样品的特征吸收光谱图,以保留时间定性,外标法定量分析。结果5种标准品的最大吸收波长分别为安赛蜜225.89 nm、苯甲酸226.87 nm、山梨酸254.97 nm、糖精钠268.68 nm、脱氢乙酸232.45 nm和292.62 nm。在1μg/ml^200μg/ml浓度内具有良好的线性关系(r=0.9999),检出限为0.001 mg/g^0.005 mg/g,回收率为96%~98%,相对标准偏差为0.19%~1.0%。结论本方法利用NaOH和乙酸锌作为蛋白共沉淀剂,能有效去除样品中蛋白,提高了方法的灵敏性,能快速实现各类食品中5种添加剂的同时检测。 Objective A fast,accurate,and simple high-performance liquid chromatography was developed for the simultaneous determination of acesulfame potassium,benzoic acid(sodium),sorbic acid(potassium),dehydroacetic acid(sodium)and saccharin sodium.Methods Different pretreatment methods were selected according to the type of sample.Methanol:ammonium acetate(pH=6.8)=5∶95(volume ratio)was used as the mobile phase,and the characteristic absorption spectra of the sample were detected by diode array detector(DAD),so as to make qualitative retention time and quantitative analysis by external standard method.Results The maximum absorption wavelength of the 5 standard products were respectively 225.89 nm of aceselmi,226.87 nm of benzoic acid,254.97 nm of sorbic acid,268.68 nm of sacrogenic sodium,232.45 nm and 292.62 nm of dehydroacetic acid.In the range of 1μg/ml-200μg/ml,the five food additives showed good linearity(r=0.9999).The limits of detection were in the range of 0.001 mg/g-0.005 mg/g,the recoveries were within 96%-98%,and the relative standard deviations were within 0.19%-1.0%.Conclusion This method uses sodium hydroxide and zinc acetate as protein co-precipitating agents,which can effectively remove protein from the sample,improve the sensitivity of the method,and quickly achieve the simultaneous detection of 5 additives in various foods.
