摘要
[目的]探讨养心通脉方促心肌梗死后大鼠缺血心肌血管生成作用及对VEGF、bFGFmRNA表达的影响。[方法]120只大鼠制成急性心肌梗死(AMI)模型,随机分为A、B、C、D、E、F 6组,每组20只;治疗6周后处死动物,取心肌组织,用免疫组织化学SP法染色组织切片测定大鼠边缘区心肌微血管数(MVC)、微血管密度(MVD)。用RTPCR法检测心肌血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(b FGF)的mRNA表达。[结果]养心通脉方各剂量组梗死边缘区MVC、MVD、VEGF、bFGF的mRNA的表达均高于模型组(P<0.05),高、中剂量组与麝香保心丸组比较,差异有显著性意义(P<0.05);低剂量组与麝香保心丸组比较,差异无显著性意义(P>0.05)。[结论]养心通脉方能促进梗死后大鼠缺血心肌局部血管新生,对缺血心肌具有保护作用,而上调心肌梗死边缘区VEGF、bFGF的mRNA表达可能是其作用机制之一。
[Objective]To investigate the effect of Yangxin Tongmai Prescription on angiogenesis of ischemic myocardium and the expressions of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)in rats after myocardial infarction.[Methods]Totally 120 rats were made into acute myocardial infarction(AMI)model and randomly divided into A,B,C,D,E and F groups with 20 rats in each group.After 6 weeks treatment,the animals were sacrificed and the myocardial tissue was taken out.The microvessel number(MVC)and microvessel density(MVD)of the rat marginal myocardium were measured by immunohistochemical SP method.The expressions of VEGF and bFGF in myocardium were detected by RT-PCR.[Results]The expressions of MVC,MVD,VEGF and bFGF in the infarction margin of each dose group of Yangxin Tongmai Prescription was higher than that of the model group(P<0.05).There was significant difference among high and medium dose groups and Shexiang Baoxin Pill group(P<0.05).There was no significant difference between low dose group and Shexiang Baoxin Pill group(P>0.05).[Conclusion]Yangxin Tongmai Decoction can promote angiogenesis in ischemic myocardium and protect ischemic myocardium in rats after myocardial infarction.Up-regulation of the expressions of VEGF and bFGF in the marginal zone of myocardial infarction may be one of its mechanisms.
