摘要
目的观察5种黄酮、皂苷类中药促心肌梗死(MI)后大鼠缺血心肌血管新生作用及对相关生长因子表达的影响。方法雄性Wistar大鼠,结扎左冠状动脉造成急性心肌梗死(AMI)模型,随机分为丹参注射液组、葛根素组、三七总皂苷组、川芎嗪组、醋柳黄酮组、模型组(对照组),并设假手术组。检测各组大鼠缺血心肌中微血管数(MVC)、微血管密度(MVD)及血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、血小板衍生生长因子β(PDGF-β)、胰岛素样生长因子(IGF-1)蛋白的表达,用病理图像分析系统测定生长因子蛋白表达灰度值,并进行半定量分析。结果模型组、各治疗组大鼠MI边缘区MVC和MVD明显高于假手术组(P<0.05);模型组VEGF、bFGF、PDGF-β、IGF-1蛋白表达及其灰度值明显高于假手术组(P<0.05);川芎嗪组、三七总皂苷组、葛根素组、丹参注射液组VEGF、bFGF、PDGF-β蛋白表达灰度值明显高于模型组(P<0.05)。醋柳黄酮组VEGF、bFGF、PDGF-β蛋白表达灰度值低于模型组(P>0.05),IGF-1蛋白表达灰度值明显高于模型组(P<0.05)。结论三七总皂苷、川芎嗪注射液、丹参注射液、葛根素注射液有促MI后大鼠缺血心肌血管新生的作用。
Objective To compare of panax notoginseng saponins（PNS）, puerarin （Pue）, salvia miltiorrhiza injection （SMI）, total flavones of hippophae rhamnoides （TFH） and tetra methyl pyrazine （TMP） on the angiogenesis and correlative growth factor expression effects in the ischemic myocardium of the rats with myocardial infarcfion（MI）. Methods The left coronary artery of male Wistar rats were ligated to make the models of acute myocardial infarction（AMI）.All rats were randornly divided into PNS group, TFH group, Pue group, TMP group, SMI group, model group （control group） and sham operation group. Microvascular vascular count （MVC）, microvascular vascular density （MVD）, vascular endothelial growth factor （VEGF）, basic fibroblast growth factor （bFGF）, insulin - kind growth factor （IGF） and platelet derived endothelial growth factor （PDGF） in ischemic myocardium of rats with AMI were detected by immunobistochemistry assay. The values of protein expression of VEGF, bFGF, PDGF -β and IGF - 1 on ischemic myocardium were calculated and analyzed by image analysis system. Resuits MVC and MVD were higher in ischemic myocardium of rats with AMI in PNS group, Pue group, TMP group and SMI group than that in model group. There were not significantly statistics differences in TFH group compared with that in model group. The values of protein expression of VEGF, bFGF, PDGF -β and IGF - 1 in model group were significantly elevated compared with that in sham operation group. The values of protein expression of VEGF, bFGF, PDGF -β and IGF- 1 in PNS group, Pue group and TMP group were significantly elevated compared with that in model group. The values of protein expression of VEGF, bFGF and PDGF -βin SMI group were significantly elevated compared with that in model group. IGF - 1 in SMI group was lower than that in model group. The values of proteins expression of VEGF, bFGF, PDGF -β in TFH group were lower than that in model group. But there were no differences between them statistically. IGF- 1 in TFH group was higher than that model group. The difference was significant. Conclusion PNS, Pue, TMP in and SMI had the role to promote anglogenesis of the ischemic myocardium in rats with MI.
