摘要
优化蒽酮-硫酸法测定秋葵多糖的检测条件。考察蒽酮浓度、硫酸浓度、蒽酮硫酸加入量和水浴时间对测定效果的影响,利用正交优化试验确定蒽酮法测定秋葵多糖的最优测定条件。试验得最优测定条为蒽酮浓度3.0g·L-1、硫酸浓度90%、蒽酮硫酸加入量10mL、显色时间10min,在此条件下达到最优效果,精密度RSD为0.73%。结果证明在此条件下,利用蒽酮-硫酸法测定秋葵多糖含量具有操作简便、精密度高可以用于秋葵多糖含量的测定。
The detection conditions of polysaccharide in Okra by anthrone-sulfuric acid method was optimized.The effects of anthrone concentration,sulfuric acid concentration,anthrone sulphuric acid addition and water bath time were studied.The optimum detecting were obtained by orthogonal test as followings:anthrone concentration 3.0g·L-1,sulfuric acid concentration 90%,anthrone sulfuric acid amount 10mL,color development time10 minutes.Under the above conditions,the RSD was 0.73%.It shows that the method has the advantages of simple operation,good repeatability and high precision.
