大叶秦艽花环烯醚萜类成分对胶原诱导性关节炎小鼠的治疗作用及相关机制探讨 被引量:12

摘要 目的:探讨大叶秦艽花环烯醚萜类成分(GMI)对胶原诱导性关节炎(CIA)小鼠的治疗作用并探讨相关机制。方法:采用CIA小鼠模型,观察GMI组(30 mg/kg和15 mg/kg)对关节炎指数的影响; HE染色观察GMI对小鼠关节滑膜组织病理学变化的影响,ELISA法检测小鼠血清中TNF-α、IL-1β和IL-6的水平,Western blot法检测滑膜中MMP1和MMP3的蛋白表达。结果:GMI能降低CIA小鼠的关节炎指数,改善关节滑膜的组织病理学改变,抑制血清中炎性因子TNF-α、IL-1β和IL-6的分泌,降低滑膜中MMP1和MMP3的表达水平。结论:GMI对CIA小鼠有一定的治疗作用,其药理作用可能通过降低血清中炎性因子TNF-α、IL-1β和IL-6的含量,并进一步抑制MMP1和MMP3的表达水平,从而减轻软骨损伤,发挥抗RA的疗效。 Objective: To observe the therapeutical effects of iridoids from the flower of Gentiana macrophylla Pall.(GMI) in collagen-induced arthritis(CIA) in mice and explore its mechanism. Methods: CIA was established to observe the effects of GMI(30 mg/kg and 15 mg/kg) on arthritis index. HE staining was used to evaluate its effects on pathological changes in joints of mice. ELISA was used to measure the levels of TNF-α,IL-1β and IL-6 in the serum. Western blot was used to detect the expression of MMP1 and MMP3. Results: GMI significantly decreased the AI in CIA mice,improved the pathological changes in joints. ELISA showed that GMI markedly down-regulated the levels of TNF-α,IL-1β and IL-6 in the serum. Western blot test indicated that GMI notably inhibited the expression of MMP1 and MMP3. Conclusion: GMI has the therapeutical effects on CIA in mice,and this effect may be associated with the depression of inflammatory factor in the serum(TNF-α,IL-1β,IL-6) and suppression MMP1,MMP3.
