摘要
目的:探讨大叶秦艽花环烯醚萜类成分(GMI)对胶原诱导性关节炎(CIA)小鼠的治疗作用并探讨相关机制。方法:采用CIA小鼠模型,观察GMI组(30 mg/kg和15 mg/kg)对关节炎指数的影响; HE染色观察GMI对小鼠关节滑膜组织病理学变化的影响,ELISA法检测小鼠血清中TNF-α、IL-1β和IL-6的水平,Western blot法检测滑膜中MMP1和MMP3的蛋白表达。结果:GMI能降低CIA小鼠的关节炎指数,改善关节滑膜的组织病理学改变,抑制血清中炎性因子TNF-α、IL-1β和IL-6的分泌,降低滑膜中MMP1和MMP3的表达水平。结论:GMI对CIA小鼠有一定的治疗作用,其药理作用可能通过降低血清中炎性因子TNF-α、IL-1β和IL-6的含量,并进一步抑制MMP1和MMP3的表达水平,从而减轻软骨损伤,发挥抗RA的疗效。
Objective: To observe the therapeutical effects of iridoids from the flower of Gentiana macrophylla Pall.(GMI) in collagen-induced arthritis(CIA) in mice and explore its mechanism. Methods: CIA was established to observe the effects of GMI(30 mg/kg and 15 mg/kg) on arthritis index. HE staining was used to evaluate its effects on pathological changes in joints of mice. ELISA was used to measure the levels of TNF-α,IL-1β and IL-6 in the serum. Western blot was used to detect the expression of MMP1 and MMP3. Results: GMI significantly decreased the AI in CIA mice,improved the pathological changes in joints. ELISA showed that GMI markedly down-regulated the levels of TNF-α,IL-1β and IL-6 in the serum. Western blot test indicated that GMI notably inhibited the expression of MMP1 and MMP3. Conclusion: GMI has the therapeutical effects on CIA in mice,and this effect may be associated with the depression of inflammatory factor in the serum(TNF-α,IL-1β,IL-6) and suppression MMP1,MMP3.
