摘要
目的观察益气化瘀解毒法对慢性萎缩性胃炎伴异型增生模型大鼠PI3K的影响。方法 56只SPF级健康雄性Wistar大鼠随机分为空白组10只,造模组46只。空白组给予SPF级动物标准饮食喂养,持续至实验结束。造模组采用以120 mg/L的N-甲基-N'-硝基-N-亚硝基胍(MNNG)溶液灌胃,每只5 m L/kg,1次/d为主,配合自由饮用0.1%氨水溶液及进食含0.03%雷尼替丁的颗粒状饲料,三种因素联合建立慢性萎缩性胃炎伴异型增生大鼠模型。20周末造模成功后将造模组剩余的36只大鼠随机分为模型组、维酶素组、益气化瘀解毒法组各12只,均给予SPF级动物标准饮食喂养。模型组予生理盐水3 m L/kg灌胃,1次/d;维酶素组予维酶素悬浊液2 m L/kg(即维酶素0.3 g/kg)灌胃,1次/d;益气化瘀解毒法组予中药制备药液3 m L/kg(含生药9 g/kg)灌胃,1次/d。持续治疗12周后即实验第32周末处死全部大鼠。选用western blot法对各组大鼠PI3K p85、pPI3K p85蛋白表达进行检测。结果各组PI3K p85蛋白表达无明显差异。p-PI3K p85蛋白表达比较,模型组表达最高,其次是维酶素组,益气化瘀解毒法组较前2组蛋白表达降低,空白组表达最少;各组灰度值比较,模型组和西药组均较空白组显著增高,模型组和西药组均较中药组显著增高,且模型组较西药组显著增高,P值均为0.000,中药组和空白组比较无统计学差异,P=0.065。结论益气化瘀解毒法可显著抑制p-PI3K蛋白的高表达,这可能是其治疗慢性萎缩性胃炎(CAG)伴异型增生(Dys)、逆转胃癌前病变(PLGC)、防治胃癌的有效作用机制之一。
Objective To observe the influence of Yiqi Huayu Jiedu principle on PI3K in rats with chronic atrophic gastritis complicating dysplasia.Methods 56 Wistar rats were randomly divided into model group（46 rats） and blank group（10 rats）.The blank group rats were given standard diet of SPF animals until the end of the experiment.Three factors were used to make model of chronic atrophic gastritis with dysplasia for the making model group rats,mainly by 120 mg/L MNNG,5 m L/kg,1 time/d,assisted by 0.1% ammonia for drinking water and 0.03% ranitidine in feed.After the model was succeeded on the20 th weekend,residual 36 rats in the making model group were randomly divided into model group,Weimeisu group and Yiqi Huayu Jiedu group,including 12 rats in each group.Each group was given the corresponding medicine once daily： 3 m L/kg saline for the model group,2 m L/kg Weimeisu suspension liquid（0.3 g/kg Weimeisu） for the Weimeisu group and 3 m L/kg herbal Solution（containing 9 g/kg crude drug）.All the rats were sacrificed after treatment for 12 weeks.The method of Western blot was used for detecting protein expression of PI3K p85、p-PI3K p85 in rats of all groups.Results There was no difference about PI3K p85 in every group（P〉0.05）.About p-PI3K p85,the protein expression of the model group and the Weimeisu group were significantly higher than the blank group（P = 0.000）; Yiqi Huayu Jiedu principle group was significantly lower than the model group and the Weimeisu group（P =0.000）; compared with the blank group,they had no difference（P = 0.065）; the Weimeisu group was also significantly lower than the model group（P = 0.000）.Conclusion Prescription of Yiqi Huayu Jiedu principle can reverse gastric mucosal dysplasia.Its effective mechanism may be related with reducing high expression of p-PI3K protein.
