神经节苷脂联合神经生长因子对脑瘫幼鼠神经行为学的影响 被引量:5

Effect of ganglioside combined with nerve growth factor on neurobehavior of infant rats with cerebral palsy

摘要 目的探讨神经节苷脂(GM1)联合神经生长因子(NGF)对脑瘫幼鼠神经行为学的影响。方法制备缺血缺氧性脑瘫幼鼠模型,随机分为模型对照组、GM1组、NGF组和联合组,每组10只;另取10只作为假手术组。联合组腹腔注射GM1 10ml/kg和NGF 10ml/kg,GM1组腹腔注射GM1 10ml/kg和生理盐水10ml/kg,NGF组腹腔注射NGF 10ml/kg和生理盐水10ml/kg,模型对照组腹腔注射生理盐水20ml/kg;每天1次,共14d。给药完成后对每组进行神经行为学评价,并检测各组大鼠血清中丙二醛(MDA)、一氧化氮(NO)和超氧化物歧化酶(SOD)的含量以及缺血侧海马区细胞凋亡情况和神经元特异性烯醇化酶(NSE)水平。结果联合组、GM1组和NGF组神经行为学实验评分均高于模型对照组(P<0.05),且联合组高于GM1组和NGF组(P<0.05)。联合组、GM1组和NGF组凋亡细胞数目、MDA和NO水平低于模型对照组(P<0.05),NSE和SOD水平高于模型对照组(P<0.05),且联合组较GM1组和NGF组更明显(P<0.05)。结论 GM1联合NGF可有效减少脑损伤造成的神经细胞凋亡,改善脑瘫幼鼠的神经行为学能力和记忆力,效果优于单一药物。 Objective To study the effect of ganglioside(GM1)combined with nerve growth factor(NGF)on the neurobehavior of infant rats with cerebral palsy.Methods Hypoxic-ischemic cerebral palsy infant rats models were established and randomly divided into four groups with 10 rats each.Another 10 rats were taken as sham-operated rats(group S).Group GM1 was injected intraperitoneally GM1 10 mg/kg and saline 10 ml/kg,group NGF was injected intraperitoneally NGF10 ml/kg and saline 10 ml/kg,group GN was intraperitoneally injected GM1 10 mg/kg and NGF10 ml/kg and group C was intraperitoneally injected the saline 20 ml/kg,once a day for 14 days.After treatment,the neurobehavior was evaluated,serum levels of malondialdehyde(MDA),nitric oxide(NO)and superoxide dismutase(SOD)were detected,and the nerve cell apoptosis and neuronspecific enolase(NSE)in the hippocampus were examined as well.Results Compared with group C,the score of neurobehavior was higher in groups of GN,GM1 and NGF(P〈0.05),which was higher in group GN than that in groups of GM1 and NGF(P〈0.05).The number of nerve cell apoptosis and levels of MOD and NO were lower,and levels of NSE and SOD were higher in groups of GN,GM1 and NGF than those in group C(P〈0.05),which were more obvious in group GN than those in groups of GM1 and NGF(P〈0.05).Conclusion GM1 combined with NGF could inhibit the nerve cell apoptosis induced by brain injury and improve the neurobehavior and memory of infant rats with cerebral palsy,which is better than the single therapy of GM1 or NGF.
