神经干细胞侧脑室内移植对脑瘫鼠的治疗效果 被引量:3

Therapeutic Effect of Lateral Ventricle Transplantation of NSCs in Rats with Cerebral Palsy

摘要 目的探讨脑瘫鼠模型采用侧脑室内神经干细胞(NSCs)移植治疗的效果。方法准备60只7日龄健康、SPF级Wistar大鼠,随机选取12只作为正常组(等量生理盐水处理),其余48只建立脑瘫鼠模型,选取24只建模成功脑瘫鼠随机分为模型组(等量生理盐水处理)和NSCs组(侧脑室内NSCs移植治疗),对比42日龄后各组大鼠的神经行为学检测结果、脑室旁白质中胶质纤维酸性蛋白(GFAP)含量差异。结果 42日龄时,模型组大鼠的Morris水迷宫逃避潜伏时间、脑室旁白质中GFAP阳性细胞数显著高于NSCs组和正常组大鼠,差异具有统计学意义(P<0.05);NSCs组大鼠的Morris水迷宫逃避潜伏时间、脑室旁白质中GFAP阳性细胞数显著的高于正常组大鼠,差异具有统计学意义(P<0.05);42日龄时,模型组大鼠的斜坡实验时间显著长于NSCs组和正常组大鼠,悬吊试验和旷场试验评分显著低于NSCs组和正常组大鼠,差异具有统计学意义(P<0.05);NSCs组大鼠斜坡实验时间显著长于正常组大鼠,悬吊试验和旷场试验评分显著低于正常组大鼠,差异具有统计学意义(P<0.05)。结论脑瘫鼠模型采用侧脑室内NSCs移植治疗能够显著改善神经行为及学习能力。 Objective To investigate the therapeutic effect of lateral ventricle transplantation of neural stem cells(NSCs)in rats with cerebral palsy.Methods Sixty 7-day-old,SPF grade wistar rats were selected.Twelve rats were selected randomly as the normal group(saline treatment),the other 48 rats were established as cerebral palsy rat model.Twenty-four successful modeling rats were randomly divided into model group(saline treated)and NSCs group(NSCs transplantation therapy).The neurobehavioral school test results and periventricular white matter glial fibrillary acidic protein(GFAP)content of rats after 42 days were compared.ResultsAt 42 days of age,Morris water maze escape latency and periventricular white matter in GFAP positive cells of model group was significantly higher than that of NSCs group and the normal group(P〈0.05).Morris water maze escape latency and periventricular white matter in GFAP positive cells of NSCs group was significantly higher than that of normal rats(P〈0.05).At 42 days of age,the slope experiment time of model group was significantly longer than that of NSCs group and normal group,while hanging test and open-field test scores were significantly lower than the NSCs group and the normal group(P〈0.05).The slopeexperiment time of NSCs group was significantly longer than that of normal group,while hanging test and open-field test scores were significantly lower than that of normal group(P〈0.05).Conclusion The lateral cerebral ventricle transplantation of NSCs can significantly improve neurobehavioral and learning ability in rats with cerebral palsy.
