螺内酯对肝纤维化大鼠Ⅰ/Ⅲ型胶原蛋白表达的影响 被引量:33

摘要 目的探讨螺内酯对实验性大鼠肝纤维化Ⅰ/Ⅲ型胶原蛋白表达的影响,初步揭示螺内酯对大鼠肝纤维化的预防作用及可能的机制.方法雄性 SD 大鼠81只,随机分为正常对照组(10只)、肝纤维化模型对照组(45只)及螺内酯组(26只).采用复合因素造模,螺内酯从造模开始每日1mL 100mg·kg^(-1)灌胃,共7wk.免疫组化染色及图像分析检测Ⅰ/Ⅲ型胶原含量.结果螺内酯组与肝纤维化组胶原面积比Ⅰ型胶原分别为:2.84±0.86,5.41±2.08,Ⅲ型胶原分别为:2.95±0.82,5.35±2.30.螺内酯组Ⅰ/Ⅲ型胶原增生程度明显降低(P<0.01).结论螺内酯可使肝组织Ⅰ/Ⅲ型胶原沉积明显减轻,提示螺内酯可明显预防肝纤维化的形成,ALD 在肝纤维化的发病中有一定意义. AIM To investigate the effects of spironolactone on expression of type Ⅰ/Ⅲ collagen proteins in experimental hepatic fibrosie In rate. METHODS Eighty one male Sprague-Dawly Rate were randomly divided into three groups,including normal control group,hepatic fibrosis model group and splronolactone group.Rats liver fibrosis was induced by compound agents(carbon tetrachloride intoxicated,high cholesterol and low protein)for a period of 7 wk. Spironolactone was given 100mg-kg^(-1).day(-1) by injecting into stomach to spironolactone group from the beginning of experiment.The expression of type Ⅰ/Ⅱ collagen were determined by immunohistochemical staining and analyzed by computer. RESULTS The results of collagen-image analysis of immunohletochemical staining show that the proliferative degree of type Ⅰ/Ⅲ collagen was significantly decresced in spironolactone group(type Ⅰ collagen in spironolactone group and hepatic fibrosis model group were 2.84±0.86 and 5.41±2.08 respectively,P<0.01; type Ⅲ collagen in spironolactone group and hepatic fibrosis model group were 2.95±0.82 and 5.35±2.30 respectively,P<0.01). CONCLUSIONS Spironolactone can significantly reduce the expression of type Ⅰ/Ⅲ collagen proteins in hepatic fibrogenesis rats.Aldosterone may play a role in hepatic fibrogenesis.
