黄芪鸡血藤汤对大鼠肝纤维化的预防作用 被引量:5

Preventive effect and mechanism of Huangqijixueteng decoction on hepatic fibrosis in rats

摘要 [目的]观察黄芪鸡血藤汤对大鼠肝纤维化的预防作用,并探讨其机制。[方法]选用SD大鼠58只,随机分为5组,正常对照组(10只),正常饮食饮水,皮下注射花生油。模型组(12只):按复合因素造模。黄芪鸡血藤汤治疗组分为大、中、小剂量组(每组12只):在造模的同时每日分别给予中药粉剂2、10、.5 g/100 g体重灌胃。8周末,光镜下观察各组大鼠肝脏组织学改变,并进行比较。免疫组化法观察黄芪鸡血藤汤对各组大鼠肝脏转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)、血小板衍生生长因子(PDGF-BB)表达的影响。[结果]8周后,中药各剂量组大鼠肝脏色泽、质地、表面光滑程度均明显优于模型组;肝细胞损伤及肝纤维化增生程度明显低于模型组(P<0.05),大剂量组最低(P<0.01);TGF-β1、CTGF、PDGF-BB在大鼠肝组织中的表达明显低于模型组(P<0.05),大剂量组最低(P<0.01)。[结论]黄芪鸡血藤汤能有效的预防大鼠肝纤维化的形成,它可能通过降低TGF-β1、CTGF、PDGF-BB的表达,抑制星状细胞的活化而起作用。 [Objective]To investigate the preventive effect of Huangqijixueteng decoction on hepatic fibrosis in rats and explore the possible mechanism. [Methods] Fiftyeight SD rats were randomly divided into five groups which include control group, the hepatic fibrosis model group and three treatment groups of Huangqijixueteng decoction (high doses group, medium doses, low doses group). Control group: the rats were injected with peanut oil 0. 3 ml/100 g subcutaneously, fed hy normal food. Model.group (twelve rats): liver fibrosis was induced by compound factors. The treatment groups (twelve rats in each group): the rats were administrated with the same way of model group, at the same time, taken Chinese herb. high dose group(2 g/100 g body weight), medium dose group(1 g/100 g body weight), low dose group(0. 5 ml/100 g body weight). At the eighth week, all rats were sacrificed. Liver tissues were stained with HE. Tissue morphological change was examined with microscopy and the degree of hepatic fibrosis was assessed by image analyze system. The expression of TGF-β1, CTGF, PDGF-BB in hepatic tissue were detected with immunohistochemical methods. [Results] Huangqijixueteng decoction could improve hepatic function. The pathology histological changes of Huangqijixueteng decoction group were significantly improverd , compared with that of the model group (P〈0. 05) and the degree of fibrosis was ameliorated(P〈0. 05). The expression of TGF-β1, CTGF, PDGF-BB in hepatic tissue were lower than those of the model group(P〈0. 05), the effects of the high dose group was surpass(P〈0. 01). [Conclusion] Huangqijixueteng decoction can improve hepatic function, prevent hepatic fibrosis in rats. This decoction perhaps plays the role through decreasing the expression of TGF-β1,CTGF, PDGF-BB in hepatic tissue to inhibit the activation of hepatic stellate cell.
