摘要
以天麻提取物为原料,采用硅胶柱层析和制备型高效液相色谱建立了分离纯化天麻素单体的制备工艺。首先经柱层析对原料进行纯化,得到天麻素粗品。通过分析型高效液相色谱考察流动相的组成、流速和上样量等对分离效果的影响,再将分析型高效液相色谱条件线性放大,确定制备型高效液相色谱条件为:流动相为乙腈—水5∶95(V/V),流速为15 mL/min,上样量为45 mg。结果显示,在该工艺条件下制备的天麻素,其纯度达到99%以上。
The preparation and purification process of gastrodin monomer from Gastrodia elata extract was established by silica gel column chromatography and high performance preparative chromatography (HPPC). The crude gastrodin was obtained from the raw material by silicone column chromatography, subsequently, the separation condition of high performance liquid chromatography (HPLC) was investigated from mobile phase composition, flow rate and sample loading capacity, finally the conditions of HPLC was linearly amplified. The HPPC conditions were established as follows: mobile phase of acetonitrile -water (5:95), flow rate 15 mL/ min, sample loading 45 rag. The results showed that the purity of gastrodin was higher than 99%.
