摘要
目的 :改进 2 0 0 0年版《中国药典》天麻中天麻素的含量测定方法。方法 :样品用 5 0 %乙醇回流提取 ;Hypersil C18色谱柱 ,流动相乙腈 水 (3∶97) ,流速 1mL·min-1,检测波长 2 2 0nm ,室温下进行测定。结果 :天麻素线性范围为 0 .0 4 0～ 4 .0 μg(r =0 .9999) ,平均加样回收率为 97.1% ,RSD为 0 .87% (n =9)。 结论 :采用 2 0 0 0年版《中国药典》天麻中天麻素的含量测定方法 ,天麻素提取不完全 ,检测灵敏度较低。本方法灵敏、结果准确可靠。
Objective: To improve the method of China pharmacopoeia(2000) for determining gastrodin in tuber of Gastrodia elata. Method: The sample was extracted by 50% alcohol. Hypersil C 18 column was used, the mobil phase was a mixture of acetonitrile water(3∶97). Flow rate was 1 mL·min -1 , the detection wavelength was 220 nm and column temperature was room temperature. Result: The calibration curve was linear (r=0.999 9) in the range of 0.040～4.0 μg for gastrodin, the average recovery was 97.1% with RSD 0.87% (n=9). Conclusion: The method was more accurate and sensitive than that in China pharmacopoeia(2000).
