摘要
目的探讨中药成分青心酮(3,4-dihydroxyacetophenone,DHAP)对RAW264.7细胞炎症反应时可溶性Toll样受体4(soluble Toll-like receptor 4,sTLR4)mRNA、蛋白表达及TNFα-分泌的影响。方法建立细胞炎症反应模型,用RT-PCR及Western blot分析sTLR4 mRNA及蛋白水平的改变,用ELISA方法检测TNFα-分泌的变化。结果RAW264.7细胞炎症反应时,sTLR4 mRNA及蛋白表达均下调,该细胞经青心酮处理后在炎症反应时,sTLR4 mRNA及蛋白表达均上调,与未处理组比较,表达明显增加,而TNFα-分泌下降。结论DHAP预处理能显著上调sTLR4 mRNA及蛋白表达,减少TNFα-分泌,抑制炎症反应。这将为临床治疗感染性休克提供一个新的研究方向。
Objective To explore the effect of DHAP on the expression of soluble Toll-like receptor 4(sTLR4) mRNA, protein and secretion of TNF-α in RAW264.7 cell line. Methods Inflammation model was established. Changes of sTLR4 mRNA and protein were analyzed by RT-PCR and Western blot. Changes of TNF-α was detected by ELISA. Results During inflanunation, the level of sTLR4 mRNA and protein was decreased, after 4h stimulated hy LPS;the level of sTLR4 protein was increased, and secretion of TNF-α was decreased when the cell line was pretreated with DHAP. Conclusion The expression of sTLR4 could be significantly up-regulated in pretreated RAW264.7 cell line with DHAP, and TNF-α secretion could be decreased. These may provide a new therapy for inflammation shock in clinical practice.
