头孢哌酮钠联合左氧氟沙星治疗对大鼠创伤弧菌脓毒症血中mCD14的影响 被引量:9

Effects of antibacterial agents on the expression of lipopolysaccharide receptor mCD14 in peripheral blood of the rats with vibrio vulnificus sepsis

摘要 目的探讨头孢哌酮钠与乳酸左氧氟沙星联用对大鼠创伤弧菌(V ibrio vu ln ificus,VV)脓毒症外周血中膜性脂多糖(LPS)受体mCD14的影响。方法制作大鼠VV脓毒症模型(VV组)和VV脓毒症药物干预模型(AA组),用兔抗鼠CD14抗体和异硫氢酸荧光素(FITC)标记的羊抗兔抗体对外周血细胞进行免疫染色,流式细胞仪测定各组FITC-CD14阳性细胞数及其荧光强度,同时观察各组染菌大鼠行为学改变情况。结果VV组FITC-CD14细胞阳性数在染菌6 h后明显升高(P<0.01),荧光强度增强;AA组在染菌6 h时,FITC-CD14细胞阳性数明显升高(P<0.01),但药物干预后FITC-CD14细胞阳性数开始减低,至染菌后12 h FITC-CD14细胞阳性细胞数为(10.967±3.135)%(P>0.05);与VV组相比,AA组在染菌后12 h和16 h的FITC-CD14细胞阳性细胞数明显减低(P<0.01)。VV组染菌4 h,大鼠出现呼吸急促,皮温升高,下肢肿胀,且症状进行性加重,染菌12 h后大鼠出现紫绀、间断抽搐等;AA组在药物干预后上述症状逐渐减轻至消失,未见抽搐,大鼠均存活。结论创伤弧菌脓毒症血中mCD14的表达水平随脓毒症的进展而升高,头孢哌酮钠联用乳酸左氧氟沙星时血中mCD14的表达水平明显减低且改善临床症状,有明显的治疗作用。 Objective To detect the effects of antibacterial agents on the expression of lipepolysaecharide ( LPS) receptor mCD14 in peripheral blood of the rats with vibrio vulnifieus sepsis. Methods To make vibrio vulnifieus sepsis model ( VV groups ) and drug intervention model(AA groups) in rats,the cells in peripheral blood were stained by immunofluoreseent method(CD14 rabbit polyelonal antibedy and FIuoreseein isothiocyanate( FITC) -conjugated goat anti -rabbit immunoglobulin specific pelyclonal antibody) and analyzed with flow eytometry( FCM ). The percentage and mean fluorescence intensity (FI) of FITC- CD14 positive cells were taken as the indexes. Meanwhile, the change in ethology was observed. Results FITC - CD14 positive cells significantly increased after 6h in VV groups (P 〈0.01 ) , and the fluorescence intensity also strengthened. FITC -CD14 postive cells of rats in AA groups increased significantly at 6h, however, it reduced after treating with antimicrobial agents. FITC - CD14 pestive cells were ( 10. 967 ± 3. 135 ) % at 12 h after being injected W ( P 〉 0.05 ) ;Compared with VV groups, FITC - CD14 postive cells at 12h and 16h after being injected VV reduced greatly in AA groups ( P 〈0.01 ). The rats in VV groups showed breathlessness, ferveseenee,and swelling in extremitas inferior at 4h after being injected VV. These symptoms were worse and worse,and the rats displayed eyanosis and convulsion at 12h; These sympetms in AA groups relieved after treating with antibacterial agents, there was no convulsion. Conclusion Our data suggest that mCD14 elevation is associated with the development of vibrio vulnifieus sepsis. The treat with Cefoperazone sodium and Levofloxaein lactate may reduced the expression of mCD14 in peripheral blood of the rats with vibrio vulnificus sepsis, and relieved the clinical symptoms.
