摘要
目的探讨抗菌药物对酒精性肝病大鼠创伤弧菌（VV）所致脓毒症肝组织Toll样受体（TLR）和促炎性细胞因子的影响。方法将制成酒精性肝病模型的66只SD大鼠随机分为酒精性肝病对照组（A组）、酒精性肝病抗菌药物对照组（AA组）、酒精性肝病VV脓毒症模型组（AV组，共4组）和酒精性肝病VV脓毒症抗菌药物治疗模型组（AVA组，共5组），并设正常对照组（N组），同时采用RT—PCR技术检测各组大鼠不同时点肝组织内TLR4、髓样分化蛋白-2（MD-2）、IL-1P和IL-6mRNA表达及其动态变化。结果AV组大鼠在染茵后各时点肝组织TLR4、MD-2、IL-1P和IL-6mRNA表达水平均较N组和A组升高（均P〈0.05或0．01），AVA组肝组织TLR4、MD-2、IL-1B和IL-6mRNA表达水平均较同时点AV组降低（均P〈0．05或0．01）。结论酒精性肝病大鼠VV脓毒症肝组织TLR4、MD-2、IL-1P和IL-6mRNA表达水平均随脓毒症病情进展而增高，而应用足量抗菌药物则可通过显著降低肝组织TLR4、MD-2、IL-1P和IL-6mRNA的表达水平，从而对酒精性肝病VV脓毒症大鼠发挥明显的治疗作用。
Objective TO detect effect of antimicrobial agents on the Toll like receptor and inflammatory cytokines in liver tissue of the alcohol-induced liver disease in rats with vibrio vulnificus sepsis. Methods Sprague-Dawley rats were randomly divided into normal control group （N group）, alcohol-induced liver disease control group （A group）, drug intervention on alcohol-induced liver disease control group （AA group）, alcohol-induced liver disease in rats with Vibrio vulnificus sepsis group （AV group）, as well as drug intervention on alcohol-induced liver disease in rats with Vibrio vulnificus sepsis group （AVA group）. To measure the expressions and dynamic changes of TLR4,MD-2,1L-1β, IL-6mRNA by RT-PCR in liver tissue of each group. Results compared with N group and A group, the expressions of TLR4,MD-2,1L-1β and IL-6mRNA in liver increased markedly in AV group （P〈0.05 or P〈0.01 ）. Compared with AV group,the expressions of TLR4,MD-2,1L-1β and IL-6mRNA in liver diminished greatly in AVA group（ P〈0.05 or P〈0.01 ）. Conclusion The expressions of TLR4,MD-2,1L-1β,IL-6 mRNA in liver tissue of the alcohol-induced liver disease in rats with vibrio vulnificus sepsis are associated with the development of vibrie vulnificus sepsis.The treat with antimicrobial agents may reduce the expressions of TLR4,MD-2,1L-1β ,IL-6mRNA in liver tissue of the alcohol-induced liver disease in rats with vibrio vulnificus sepsis,and have the visible therapeutical effect for alcohol-induced liver disease in rats with vibrio vulnificus sepsis.
