摘要
以水稻、玉米为材料 ,对其基因组DNA的提取方法进行改进 ,通过减少饱和酚和氯仿等有机试剂处理次数 ,适当延长有机试剂的抽提时间 ,提高提取DNA的得率和纯度 ,并且能更有效地避免干扰物质的污染 ,得到均一的大片段的DNA。检测结果表明 ,该方法普遍适合玉米、水稻作物材料的DNA提取 。
In this paper, a modified method for DNA extraction and purification was described. The more purified and heavier DNA molecule could be obtained by properly decreasing the extraction times with the organic solvent such as phenol-chloroform and increasing the treating time with the organic solvent. The result showed that this modified method was appropriate for the DNA (extraction) of the maize and rice, and the purified DNA could be met the desired requirement of further genomic research as well.
