摘要
目的:对川芎内生菌Peniophora incarnata代谢产物的化学成分进行研究。方法:用糙米进行固体发酵获得P.incarnata菌的代谢产物,对其乙酸乙酯部位采用大孔树脂柱、中压C18柱、半制备液相等方法分离得到单体化合物,再根据核磁共振结果鉴定得到这些化合物的结构。结果:分离得到22个化合物,分别为吡咯并哌嗪-2,5-二酮(1)、环-(脯氨酸-苏氨酸)(2)、环-(L-丙氨酸-L-脯氨酸)(3)、环-(D-丙氨酸-L-脯氨酸)(4)、环-(L-酪氨酸-L-脯氨酸)(5)、环-(苯丙氨酸-脯氨酸)(6)、环-(苯丙氨酸-缬氨酸)(7)、环-(亮氨酸-亮氨酸)(8)、环-(L-异亮氨酸-L-亮氨酸)(9)、阿魏酸甲酯(10)、反-对香豆酸甲酯(11)、(1H-indol-3-ly)oxoacetamide(12)、1H-indole-3-carboxaldehyde(13)、indole-3-acetic acid methyl ester(14)、1H-indole 3-carboxylic acid methyl ester(15)、对羟基苯甲醛(16)、对羟基苯甲酸甲酯(17)、胸苷(18)、oryzalactam(19)、肌苷(20)、(3R,12aS)-3-methyl-2,3,6,7,12,12a-hexahydropyrazino[1,2-b]β-carboline-1,4-dione(21)、(3S,12aS)-3-(1’-Methylpropyl)-2,3,6,7,12,12a-hexahydropyrazino[1’,2’:1,6]pyrido[3,4-b]indole-1,4-dione(22)。结论:本研究分离得到的单体化合物主要为环二肽类化合物,其中化合物1~22首次从该种真菌的次生代谢产物中分离得到。
Objective:To study the chemical composition of the metabolites of endophytic fungus Peniophora incarnata of Chuanxiong.Method:The metabolites of P.incarnata bacteria were obtained by the solid fermentation of brown rice.The ethyl acetate part was separated by the methods of macroporous resin column,medium pressure C18 column and semi-preparative liquid phase to obtain monomer compounds,and their structures were identified according to NMR results.Result:22 compounds were isolated,including pyrrolopiperazine-2,5-dione(1),cyclo-(Pro-Thr)(2),cyclo-(L-Ala-L-Pro)(3),cyclo-(D-Ala-L-Pro)(4),cyclo-(L-Tyr-L-Pro)(5),cyclo-(Phe-Pro)(6),cylco-(Phe-Val)(7),cyclo-(Leu-Leu)(8),cyclo-(L-isoleu-L-leu)(9),4-Hydroxy-3-methoxycinnamic acid methyl ester(10),trans-coumaric acid methyl ester(11),(1H-indol-3-ly)oxoacetamide(12),1H-indole-3-carboxaldehyde(13),indole-3-acetic acid methyl ester(14),1H-indole 3-carboxylic acid methyl ester(15),4-hydroxybenzaldehyde(16),methyl 4-hydroxybenzoate(17),thymidine(18),oryzalactam(19),inosine(20),(3R,12aS)-3-methyl-2,3,6,7,12,12ahexahydropyrazino[1,2-b]β-carboline-1,4-dione(21),(3S,12aS)-3-(1’-Methylpropyl)-2,3,6,7,12,12a-hexahydropyrazino[1’,2’:1,6]pyrido[3,4-b]indole-1,4-dione(22).Conclusion:The monomeric compounds isolated in this study are mainly cyclic dipeptide compounds,and the compounds 1~22 are isolated from the secondary metabolites of the fungus for the first time.
