摘要
目的:探讨柴胡三参胶囊对缺血性心律失常大鼠心肌组织微小核糖核酸-29(Microribonucleic Acid-29,miRNA-29)表达的影响。方法:将40只Wistar大鼠随机分为空白组、对照组、柴胡三参胶囊组、维拉帕米组,每组10只。干预10天后,除空白组外,其余各组采取大鼠左冠状动脉前降支高位结扎法制作缺血性心律失常模型,记录各组大鼠室性早搏(Ventricular Premature Beats,VPB)次数以及首次出现时间,室性心动过速(Ventricular Tachycardia,VT)和心室纤维性颤动(Ventricular Fibrillation,VF)次数和持续时间,计算各组大鼠心律失常评分;qRT-PCR检测各组大鼠心肌组织miRNA-29的表达。结果:与空白组相比,其他三组出现明显的缺血性心律失常,VPB、VT和VF频率明显增加(P<0.01),VPB首次出现时间缩短(P<0.05),VT和VF持续时间增长(P<0.05),miRNA-29表达升高(P<0.05);与对照组相比,柴胡三参胶囊组、维拉帕米组VPB、VT和VF频率明显降低(P<0.05);VPB首次出现时间推迟(P<0.05),VT和VF持续时间也降低(P<0.05),心律失常评分降低(P<0.05);心肌组织miRNA-29表达更低(P<0.05)。结论:柴胡三参胶囊可以降低心肌组织中miRNA-29的表达水平,减少缺血性心律失常的发生。
Objective:To explore the effects of the Chaihu Sanshen capsules(柴胡三参胶囊)on the expression of miRNA-29 in myocardial tissues of ischemic arrhythmia rats.Methods:A total of 60 Wistar rats were randomly divided into the blank group,the control group,the Chaihu Sanshen capsules group and the verapamil group,10 rats in each group.After corresponding intervention for 10 days,the blank group was left without any treatment,and the rats in the other 3 groups received high-level ligation at the left anterior descending coronary artery to make an ischemic arrhythmia model.Results:Compared with the blank group,the other 3 groups showed more obvious ischemic arrhythmia,the frequencies of VPB,VT and VF were increased significantly(P<0.01),and the first time of VPB appeared was significantly shortened(P<0.05).The duration of VF and VF was increased(P<0.05).The expression of miRNA-29 in myocardial was increased(P<0.05).Compared with the control group,the frequencies of VPB,VT and VF in the Chaihu Sanshen capsules group and the verapamil group were obviously decreased(P<0.05),the first time of VPB appeared was delayed(P<0.05),the duration of VT and VF was shortened(P<0.05),the arrhythmia score was decreased,and the expression of myocardial miRNA-29 was decreased(P<0.05).Conclusion:The Chaihu Sanshen capsules can decrease the expression of miRNA-29 in myocardial tissues of ischemic arrhythmia rats,thereby reducing the occurrence of ischemic arrhythmia.
